Objective:Exosomes are small-sized vesicles, which are secretory extracellular vesicles released from cells into the circulation system.Exosomes always carry mounts of major biological information and play important roles in multiple complicated biological processes, such as regulating cell cycle, cell proliferation, cell apoptosis, cell aging and so on.As for cardiovascular system, exosomes participate in the processes such as cell survival, cardiac regeneration, ventricular remodeling and angiogenesis. In this study,we are trying to make clear what roles the exosomes play in the apoptosis process of rat H9C2 cardiomyocytes which is induced by H2O2.Study methods and results:1.Using western bolt,we found that the exosomes from plasma were positive immunoreactivity to exosome surface marker proteins CD63.2.Flow cytometry results showed that exosomes decreased H2O2-induced apoptosis cells numbers and improved survival in rat H9C2 cardiomyocytes.Western blot analysis showed increased BCL2 level, decreased BAX level and decreased cleaved caspase3/caspase3 level in exosomes-treated groups. Conclusions:Theses data indicated that plasma exosomes have protective effects against rat myocyte apoptosis induced by H2O2.
