Study On The Effect Of Kidney-Reinforcing Herbal Medicine On Aged Infertility Women's Fertility Based On Proteomics

Objective:Studing and identifying the follicular fluid proteome differential protein in the elderly and young women,and the both kidney Qi and Yin deficiency elderly women,searching for the biological function of these proteins,from the microscopic,verify the hypothesis:kidney deficiency caused by "57" is the symptoms of both kidney Qi and Yin deficiency.Observe the effect of the traditional Chinese medicine for tonifying kidney and benefiting Yin erzhi tiangui recipe on the outcome of IVF-ET in the both kidney Qi and Yin deficiency elderly women and the biological mechanism of follicular fluid proteome.Methods:50 cases were selected in the young blank group,and the other in the elderly blank group(n = 50).By using the random number and the envelope method,100 both kidney Qi and Yin deficiency elderly women cases were divided into two groups:the control group and the aged group.These 100 cases were given placebo or erzhi tiangui recipe combined with antagonist regimen on the day of starting OS treatment.Observe the difference of the groups:syndrome score before and after treatment,the Gn days,Gn amount,the number of oocytes>2PN and cleavage,the number of grade I embryos and grade(? + ?)embryos.Identify the differentially expressed proteins by LC-MS/MS technique,retrieval bioinformatics function,and verify some key proteins by Western Blot.Results:Between the elderly.both kidney Qi and Yin deficiency elderly women and the young women,there were similar differences in proteomic of follicular fluid?Follicular fluid proteome showed high consistency in the elderly and both kidney Qi and Yin deficiency elderly women?In the placebo group,the number of grade I embryos and high-grade embryo rate were lower than those of erzhi tiangui recipe group(0.64±1.03vs2.11±2.05,17.1%vs51.1%),and the difference was statistically significant?No significant difference in other indicators.Compared with the erzhi tiangui recipe group and the placebo group,the effect of apoptosis in follicular fluid was down regulated,and the Western Blot experiment verified that the expression of IGFBP3 and TIMP1 were down regulated and up-regulated,respectively.Conclusion:The elderly and both kidney Qi and Yin deficiency elderly women have similar follicular fluid proteome basis.After treatment with Chinese herbs,the causation for the improvement of IVF-ET outcome is down regulation of the expression of apoptosis protein in follicular fluid?...
