Oviductus Ranae,also known as snow clams,has been regarded as one of the most important traditional Chinese medicines and nourishing health products.Pharmacological studies have shown that Oviductus Ranae has the effect of promoting growth of animals,anti-fatigue,anti-stress,enhance immunity,prevention and treatment of children with asthma,anti-tumor,anti-depression and other effects.The chemical compositions of the Oviductus Ranae are complex,so the research progress of active ingredients of Oviductus Ranae is slow.In this paper,the quality evaluation,effective chemical composition,pharmacological properties and effective chemical constituents of Oviductus Ranae were studied systematically.Oviductus Ranae is mainly produced in Changbai Mountain area of Jilin Province.It is a traditional Chinese medicine grew in Changbai Mountain area.In this paper,27 samples from main producing areas of Oviductus Ranae were collected,which covered the main source of Oviductus Ranae in China.In this paper,the quality evaluation method of Oviductus Ranae,the quality of the main producing areas,and the active constituent derivatives were systematically studied.The expansion coefficient,alcohol extract,total water content and total ash of 27 batches of Oviductus Ranae were tested in the main producing areas of Changbai Mountain.The results showed that the expansion coefficients of 27 batches of samples were different,but they all accorded with the pharmacopoeia.The results of other physical and chemical indicators for the further optimization of the origin of Oviductus Ranae processing technology and test analysis provides a reference.In addition,this paper has improved the quality control system of Oviductus Ranae by thin layer chromatography and fingerprint spectroscopy research.The research results of 1-methyl hydantoin pre-column derivatization greatly improved the 2015 version of "Chinese Pharmacopoeia" Oviductus Ranae standard chromatographic conditions,and improved the detection sensitivityIn this paper,the anti-fatigue effect of Oviductus Ranae was studied.The anti-fatigue animal model was established.The anti-fatigue activity of the petroleum ether extract of Oviductus Ranae was selected.Three main anti-fatigue and biochemical indexes,namely blood lactic acid,serum urea nitrogen and liver glycogen,and further explored the anti-fatigue mechanism of petroleum ether extract of Oviductus Ranae.Subsequently,the correlation analysis was carried out to analyze the correlation between the 19 peaks of the total peak area and the anti-fatigue ability of the petroleum ether extract of Oviductus Ranae in different batches to obtain the correlation degree.Each chromatographic peak was resistant to fatigue effect of the relationship.The gray correlation analysis showed that the chemical composition represented by each peak in the fingerprint spectra was correlated with the anti-fatigue effect(P> 0.65),indicating that the anti-fatigue effect of the petroleum ether layer was the result of the chemical composition.According to the size of the correlation degree,the order of the contribution of each component to fatigue resistance was determined as follows: 3> 2> 5> 15> 4> 17> 8,and it was pointed out that the 17 th peak was 7-keto cholesterol.In order to explore antitussive activity of Oviductus Ranae,the pharmacokinetic study the antitussive active ingredients derivatives,1-methylhydantoin ibuprofen and 1-methylhydantoin aspirin,were studied.The synthesis of 1-methylhydantoin ibuprofen and 1-methylhydantoin aspirin were synthesized by amination of 1-methylhydantoin with the carboxyl groups of ibuprofen and aspirin.The chemical structures of the synthesized 1-methylhydantoin ibuprofen and 1-methylhydantoin aspirin were characterized by proton and carbon NMR spectroscopy and single crystal structure analysis.Then we studied the anti-inflammatory and cough pharmacodynamics experiments.The results showed that 1-methylhydantoin ibuprofen,1-methylhydantoin aspirin antitussive and anti-inflammatory activity was superior to single 1-methylhydantoin and ibuprofen or aspirin alone which provided a reference for the development of anti-inflammatory and antitussive drugs...
