The clinical experience summary is an important issue that matters the development and innovation of Traditional Chinese Medicine(TCM.)As the development of the society,more and more His data are mounting,so how to adopt the clinical data efficiently to make good analyze based on that become problems to be solved.The paper aimed to make some exploration on the clinical data application and clinical experience summary,which is carried out via an example of the His data analysis based on Chief Yang Weibin.We proposed the opinion that doctor’s clinical analysis should be carried out on the new model "Follow the teacher learning should be combined with His data analysis via data mining,mechanism investigation via network pharmacology and pharmacological experiment",which would be a new pathway for clinical experience summary.And it might boost the transmitting of clinical experience.The job would be carried out from three aspects:1.Hospital His data-miningWe use software TCMISS V2.5 which is integrated with various data mining methods,such as entropy methods and association rule analysis,to make analysis on the clinical His data with 1423 clinical cases within four years.The academic idea on the rheumatoid arthritis(RA)treatment was summarized and core drug composition rules were mined out,and finally a core prescription with all the main drugs such as Cmnamomi Mmulus,Paeoniae Radix Alba,Glycyrrhizae Radix Et Rhizoma,Ephedrae Herba,Atractmlodis Macrocephalae Rhizoma,Anemarrhenae Rhizonma,Saposhnikoviae Radix,Aconiti Lateralis Radix Praeparrka,etc.were obtained.2.Core prescription optimization and mechanism explorationBased on the outcome of data-mining in Step 1,we carried out a core prescription optimization work to get an optimized prescription,and make mechanism exploration on that.Here network pharmacology was applied.Firstly,get the potential targets of the drug via similarity comparison with other marketing drugs,as well as the genes related to the RA disease.Then via String and Cytoscape to establish the PPI(Protein-Protein Interaction)network,and via topological features computing,the core genes/proteins in the network could be obtained.Next,enrichment analysis is carried out to get the main Kegg pathways related with RA treatment,including Osteoclast differentiation,TNF signaling pathway,Toll-like receptor signaling pathway,T cell receptor signaling pathway,MAPK signaling pathway,Cytokine-cytokine receptor interaction,NF-kappaB signaling pathway.3.Pharmacological experiment for mechanism validationAccording to the 8 pathways obtained in Step 2.Osteoclast differentiation pathway was chosen to make deep analysis.We use CIA model rat,with the intervention of Chinese medicine(core prescription),three doses to make the phamacodynamics study.The main evaluation index included pathological score,TRAP,Micro-CT etc.It shows that the Chinese medicine(core prescription)has good efficacy on RA treatment.Meantime,we use western blot and Elisa methods to detect the 5 protein enriched in the Osteoclast differentiation pathway,including IFNG,c-FOS,c-JUN,IFN-γ,ILl-B.And finally,all of them had showed lower expressing quantity after taking TCM,which means that the core prescription had good efficacy on CIA model and the mechanism are related with Osteoclast differentiation pathway.In a word,the article mainly focuses on to establish a new model for clinical data application,hoping to combine both traditional experience and modern techniques perfectly,which would be beneficial to TCM composition rule analysis as well as TCM clinical data application.
