Purpose: to primary screening of differential expression of serum microRNAs before and after the radiochemotherapy among the Han,Uygur and Kazak locally advanced esophageal squamous cell carcinomas(ESCC)patients;the serum miRNAs differential expression between radiochemotherapy sensitive group and resistance group to find radio-chemotherapy short-term efficacy predictive factors.Methods: collect 30 patients with locally advanced ESCC,blood samples also collected before and after radiochemotherapy,at the same time to collect normal people blood as a control.Agilent microRNA gene chip was applied to detect serum miRNA levels of each subject.The differential miRNAs were imported to DIANA-mirPath.v3 to analysis biological functions.RT-qPCR to validate differentially expressed microRNAs between sensitive and resistance group.Results: 1.the short-term efficacy follow-up result:14 cases of CR+PR in sensitive group,16 cases of SD+PD in resistance group.2.(1)compared before radiochemotherapy with nomal group:a total of 98 miRNAs expression significantly different in locally advanced ESCC;after radiochemotherapy,a total of 81 microRNAs expression different significantly;only miR-623 differently expressed before and after radiochemotherapy.(2)microRNAs expression differences more than three times were:miR-150-5p,mi R-486-5p,miR-16-5p,and miR-3185;(3)KEGG pathway analysis revealed that miR-623 may regulate the NF-kappa B signaling pathway.3.(1)A total of160 miRNAs different expression significantly in the Han nationality before radiochemotherapy group compared with normal control group;after radiochemotherapy,a total of 124 microRNAs different expression significantly;6 microRNA differentexpression between after and before group,;(2)for Uygur,total of 92 microRNAs expression different significantly before radiochemotherapy group,and 103 microRNAs in after group;1 microRNA up regulated and 1 down regulated between after and before group;(3)the Kazak has a total number of 115 microRNAs in before radiochemotherapy group,129 microRNAs in after radiochemotherapy group,4 up regulated microRNAs between after and before group;(4)the Han and Kazak patients shared 6 differentially expressed miRNAs,the Kazak and Uygur patients shared 10,while Uygur and Han patients shared 17;mi R-4454,miR-144-3p,miR-4459 obvious differentially expressed in three nationalities.4.11 microRNAs were different expression between sensitive and resistance group;miR-623?miR-5703 and miR-483-5p were differentially expressed by RT-qPCR;ROC curve used for radiochemotherapy short-term effection prediction.For miR-623 the AUC?sensitivity and specific degrees were 0.809?72.2%?82.0%,mi R-5703 were 0.928?91.7%?90.0%,mi R-483-5p,0.838?69.4%?90.0% respectively.Conclusion:serum miRNAs are differentially expressed in ESCC patients.In addition,miRNAs were different expression among three nationality groups on ESCC patients.MiR-483-5p,miR-623,miR-5703 may be radiochemotherapy short-term efficacy predictive factor for ESCC.
