Background and PurposeNowadays,esophageal cancer is a common tumor of digestive system in the world.The incidence and mortality of esophageal cancer in China are rising,which has great impact on the health level of the residents in our country.Recent studies have shown that the genetic and a variety of environmental factors may contribute to the incidence of esophageal cancer,and especially the polymorphism of interleukin(IL-10)gene can lead a person to suffer from increased susceptibility to esophageal cancer.Therefore,our study set out to analysis the relativity on three common IL-10 gene loci(-1082 G /A rs1800896,-819T/C rs1800871,and-592A/C rs1800872)with the risk of suffering from esophageal cancer to explore the correlation between the polymorphism of IL-10 gene and the susceptibility to esophageal cancer.Methods1.We used the purposive sampling,271 patients who were diagnosed with esohpagus cancer from the Second Affiliated Hospital of ZhengZhou Universityduring the period between March 2011 and May 2014,are selected as experimental group and 492 healthy people which recruited at the health examination center as control group,to conduct case-control study,in which general information is surveyed by questionnaire.2.Blood samples are collect among all the objects of the study and gene extraction is conducted,cancer tissues of 246 patients from the experimental group are researched by immunohistochemisty and Western-Blot.To the cultivation of the esophageal cancer cell lines in vitro,and treatment of IL – 10,to observe the impact of the IL-10 on the esophageal cancer cell proliferation,metastasis and invasive.3.Chi-squared test were used to analysis the difference and compare by analyzing general information of the objects between the cases and controls group and the probability of environmental carcinogenic factors contact.4.Compare the number of different genetic locus defect from the two groups and analyze it by OR.Compare the number of cases that the two study objects with different genetic locus defect that exposed to environmental carcinogenic factors as smoking,drinking and first degree relatives family history and analyze it by OR.ResultsThe demographic and clinical characteristics of all of the esophageal cancer cases and controls had no significant difference(P>0.05).Unexpectedly,our study found that the patient who had an esophageal cancer was more likely to have a not so good habit of smoking and drinking,and had a higher probability to have a family history of cancer in first relatives(P<0.05),comparing with control groups.Of 246 patients,94(38.21%)were at stage I-II,and 152(61.79%)were at stage III-IV.The genotype distributions of IL-10-1082G/A rs1800896,-819T/C rs1800871,and-592A/C rs1800872 confirmed that the controls had a HWE,and P values forHWE were 0.74,0.06 and 0.53,respectively(Table 3).Unconditional logistic regression analyses showed that subjects carrying the AA genotype and GA+AA genotype of IL-10-1082G/A rs1800896 were associated with an increased risk of esophageal cancer,and the adjusted OR(95% CI)were 2.19(1.31-3.64)and 1.44(1.05-1.99),respectively.However,we did not find significant relationship between IL-10-819T/C rs1800871 and-592A/C rs1800872 with the development of esophageal cancer.In addition,IL-10-1082G/A rs1800896 polymorphism also has no significant association with smoking,drinking and family history of cancer in the first relatives on the risk of esophageal cancer(P>0.05).ConclusionsSmoking,drinking and family history of esophageal cancer may be associated with the occurrence of esophageal cancer.Rs1800896 IL-10-1082G/A polymorphism is associated with the susceptibility to esophageal cancer,but its role has no relationship with smoking,alcohol consumption,and family history of esophageal cancer.Therefore,it is worth to wait that rs1800896 IL-10-1082G/A gene mutation is used as candidate biomarkers to predict the sensitivity of esophageal cancer for people,which is of great significance for the next step for the prevention and control of esophageal cancer.We can study IL-10-1082G/A rs1800896 polymorphism and other environmental factors(such as speed of diet and dietary categories: vegetables,fruits,eggs and milk,pickles,etc.)have no significant relationship,as well as the relationship between some gene polymorphisms.
