Sophora alopecuroides is a perennial herb growing in desert,grassland and farmland,belonging to the genus sophora.China is a great growth country of Sophora alopecuroides.More than half of the world's yield is produced by our country.It is widely distributed in northern desert areas of Ningxia,Gansu,Qinghai and Xinjiang in China,and there are about 12 types of Sophora alopecuroides.From the role of Sophara ALOPECURIDES main ingredients are alkaloids containing the.The alkaloids contained 20 kinds of alkaloids,such as matrine,Oxymatrine,Oxymatrine and matrine.The main effect of the drug is heat and dampness,detoxification,diuresis,jaundice disappear in inflammation.Modern pharmacological research confirmed that more content and medicinal value of bitter bean grass is larger,more research bases of matrine with heat,dampness,anti-inflammatory,antiviral,diuretic,anti ulcer,anti diarrhea,anti arrhythmia,analgesia cooling,lowering blood pressure and blood lipid and other medicinal value.It has been reported that the extract of Sophora alopecuroides has certain effect on the treatment of cow mastitis.However,there are few studies on the bioactivities of alkaloids from Sophora alopecuroides at home and abroad.Therefore,the pharmacological activity of total alkaloids of Sophora alopecuroides was studied in this paper.The details are as follows:1.Study on acute toxicity of total alkaloids of Sophora alopecuroidesFirst,the acute toxicity test of total alkaloids of Sophora alopecuroides were studied in this paper.The acute toxicity test was carried out by using the total alkaloids of Sophora alopecuroides in rats.The maximum lethal dose and the minimal lethal dose were determined by pre experiment.Then,four concentrations of total alkaloids of Sophora alopecuroides were prepared.After filling,the behavior changes and clinical manifestations of the rats were observed and recorded.10 d was continuously observed,and the number of dead rats was recorded in detail.Then we dissected the dead rats and examined the organs,including the subcutaneous,heart,liver,lung,spleen,brain and mesentery.Based on the simplified Karber median lethal dose(LD50),its value is 121.42mg/kg,the 90% confidence interval was105.4~120.5mg/kg,indicating acute toxicity.2.Experimental study on chronic toxicity of total alkaloids of Sophora alopecuroidesIn order to evaluate the toxicity of total alkaloids of Sophora alopecuroides,the long-term toxicity test was carried out to evaluate the safety of the drug.3 high dose,medium and low dose groups and one blank control group were set up.Wistar rats were orally administered 1 times of total alkaloids solution of Sophora alopecuroides by gavage with metallic gastric tube every day,28 d continuously,and rats in contract group were injected with pure water.By observation of the general situations and the pathological changes of main organs and the change of body weight changes,blood physiological and biochemical indexes,the results showed that there was no adverse effect,the physiological activities of total alkaloids of Sophora alopecuroides perfusion in rats,blood physiological and biochemical indexes the main organs.During the experiment,no toxic reaction occurred and no death occurred in each dose group.The experimental results showed that the total alkaloids of Sophora alopecuroides had no chronic toxic and side effects on the experimental animals.3.Study on the common pathogenic bacteria of cow mastitis with total alkaloids of Sophora alopecuroidesStaphylococcus aureus,Escherichia coli and Streptococcus lactis were the main bacteria causing mastitis.About 90% of cow mastitis is caused by the over proliferation of these three bacteria.Therefore,we carried out the experimental study on the inhibitory effect of total alkaloids of Sophora alopecuroides on the three bacteria in vitro.We choose the total alkaloids of Sophora alopecuroides in different concentrations of liquid poured into the agar and then inoculated with common bacteria that cause mastitis,minimum inhibitory concentration determination of total alkaloids of Sophora(MIC)and minimum bactericidal concentration(MBC)and PH,serum concentration and inoculation quantity of MIC.The results showed that the MIC of Staphylococcus aureus,Escherichia coli and Streptococcus lactis were 6,6and 3 mg/ml,respectively,and the average MBC were 100,100,100,50mg/ml,respectively.PH only had an obvious effect on Escherichia coli MIC,but had no obvious effect on Staphylococcus aureus and Streptococcus lactis.The inoculation amount of three kinds of bacteria increased,and the MIC value also increased.The concentration of serum in culture medium had no obvious effect on the three strains of MIC.The results showed that Streptococcus lactis was the most sensitive to the total alkaloids of Sophora alopecuroides,and the other two strains were the second.Therefore,we concluded that the total alkaloids of Sophora alopecuroides can be used as the treatment of cow mastitis.4.Effects of total alkaloids of Sophora alopecuroides on immune function in ratsTo investigate the effect of total alkaloids from Sophora alopecuroides on immune function in rats.We have to total alkaloids of Sophora alopecuroids perfusion fluid with different density of spleen cells of rats in the medium,by measuring the effect on lymphocyte proliferation MTT assay results showed that total alkaloids of Sophora alopecuroides in the 70~280 range of ug/ml inhibited the proliferation of B lymphocytes,but to promote T cells proliferation.Meanwhile,the effect of total alkaloids of Sophora alopecuroides on tumor necrosis factor(TNF-?)was detected by double antibody sandwich ELISA.The results showed that different concentrations of total alkaloids of Sophora alopecuroides inhibited the production of TNF-alpha.Then,the effect of total alkaloid of Sophora alopecuroides on the activity of interleukin 2(IL-2)in rats was detected by 3H-Td R incorporation method.It was found that the total alkaloids of Sophora alopecuroides could enhance the activity of IL-2 in the range of 70~280 ug/ml.All above experiments showed that the total alkaloids of Sophora alopecuroides infusion solution affected the immune function of rats,inhibited the proliferation of B lymphocytes and the production of TNF-alpha,promoted the proliferation of T lymphocytes and enhanced the activity of IL-2.
