Taken Guangxi Bama pigs as the object,the research screened differentia 1 genes of longissimus muscle Guangxi Bama pigs in 30,180 days of age thr ough RNA-seq and Two-dimensional Gel Electrophoresis,and revealed the mec hanism of the differences of production performance,meat quality,intramuscula r fat deposition between Guangxi Bama pigs and landrace in accordance with e nzyme activities related to muscle metabolism,biochemical and physiological in dexes,gene expression,etc.The results are as follows;1.Results of meat quality,metabolizing enzymes activity and physiological and biochemical parameters in Bama pig and Landrace:(1)Guangxi Bama pig average back-fat thickness were 4.23 ± 0.15cm,significantly higher than Landrace 1.47±0.21cm(p<0.05),average muscle conductivity were 2.80±0.49ms,significantly lower than Landrace 4.24±0.51ms(p<0.05),average Opto muscle flesh were 64.64± 2.80,higher than Landrace 63.44 ± 0.88,the difference was not significant,average tenderness were 16.13 ± 1.26 N,lower than Landrace 17.89±0.96N,the difference was not significant,average eye muscle area were 13.63 ± 0.41 cm2,significantly lower than Landrace 36.58±2.37 cm2(p<0.01)in 180 days of age.(2)Results of metabolizing enzymes activity and physiological and biochemical parameters in Guangxi Bama pig and Landrace followed the same age-dependent patterns;FFA concentrations were significantly higher than Landrace(p<0.05),LDH,TC,TG,LDL,HDL concentrations were significantly lower than Landrace(p<0.05)in 0 days of age;FFA,TG concentrations,LDH,MDH SDH activities in longissimus muscle were significantly higher than Landrace(p<0.05),TC concentrations were significantly lower than Landrace(p<0.05)in 30 days of age;FFA,TG,LDL,HDL concentrations,LPL,MDH,SDH activities in longissimus muscle were significantly higher than Landrace(p<0.05),LDH activity in longissimus muscle were significantly lower than Landrace in 180 days of age(p<0.05).(3)Guangxi Bama pig LPL,LDH,MDH,SDH activities in longissimus muscle between IMF content were no significant correlation(p>0.05).2.Results of Guangxi Bama pig and Landrace longissimus muscles RNA-seq sequencing in 30,180 days of age.(1)Total 23051730,21475968,23000120 and 18698878 clean reads were identified,After assembled,there were 27,491 unigenes obtained,19,655(71.49%)were annotated to Unigene database,7836(28.51%)Unigenes were not annotated in guangxi Bama pig.There were 69686 unigenes obtained,43447(62.34%)were annotated to Unigene database,26239(37.66%)Unigenes were not annotated in Landrace.(2)Two libraries were constructed in order to identify differential expressed genes between longissimus muscle Guangxi Bama pigs in 30,180 day of age.Transcriptome comparison revealed 1380 differentially expressed genes,1,141 of which were up-regulated,whereas 239 were down-regulated,2873 genes were assigned GO terms for biological processes,2713 genes were assigned GO terms for cellular component,323 genes were assigned GO terms for fmolecular function,a total of 206 differentially expressed genes were assigned with one or more KEGG annotation,they were mapped to 178 KEGG pathways,43,656,37,646 SNPs were respectively found of longissimus muscle Guangxi Bama mini pigs in 30,180 days of age.(3)Two libraries were constructed in order to identify differential expressed genes between longissimus muscle Landrace in 30,180 day of age.Transcriptome comparison revealed 351 differentially expressed genes,212 of which were up-regulated,whereas 139 were down-regulated,2609 genes were assigned GO terms for biological processes,1894 genes were assigned GO terms for cellular component,892 genes were assigned GO terms for fmolecular function,a total of 258 differentially expressed genes were assigned with one or more KEGG annotation,they were mapped to 126 KEGG pathways,43,656,37,646 SNPs were respectively found of longissimus muscle in Landrace 30,180 days of age.3.Results of proteome analysis of longissimus muscle Guangxi Bama pigs in 30,180 days of age and Landrace in 180 days of age performed by two-dimensional gel electrophoresis.(1)The isoelectric point of muscle protein between 4-7,the molecular weight range in 25-100 kDa,proteom comparison revealed 267 differentially expressed protein spots,149 of which were up-regulated in 30 days of age,whereas 148 were up-regulated in 180 days of age.(2)A total of 67 protein spots were identified by Mass Spectrometry,32 of which were identified in Guangxi Bama pigs 30 days of age,whereas 35 were identified in Guangxi Bama pigs 180 days of age.(3)A total of 16 protein spots were identified by Mass Spectrometry,13 of which were identified in Guangxi Bama pigs 180 days of age,whereas 3 were identified in Landrace 180 days of age.4.Gene expression of differentially expressed genes in Guangxi Bama pig and Landrace longissimus muscle.(1)The developmental changes of NDUFS3,NDUFAB1,IDH3A,NU6M,ATP5B,ATP5H,HUMMLC2B,MYL1,ADPN mRNA abundance in Guangxi Bama pig and Landrace longissimus muscle presented the same age-dependent patterns,which decreased with age(p?<0.05).(2)There were positive correlations between NDUFAB1,NU6M mRNA levels and IMF content(0.826(p<0.05)and 0.848(p<0.05)respectively),a negative correlation between ATP5H mRNA levels and IMF content(-0.894(P<0.05)),negative correlations between HUMML2CB mRNA levels and shear force,muscle conductivity,IMF content,(-0.984(p<0.01),-0.895(p<0.05),0.894(p<0.05)respectively),positive correlations between TPI mRNA levels and muscle shear force,conductivity extremely relevant,IMF content,(0.880(p<0.01),0.852(p<0.01),0.820(p<0.05)respectively),and a positive correlations between ADPN levels and shear force(0.840(p<0.05)).
