Foot-and-mouth-disease virus (FMDV) can cause animal blisters on hoof and lips, it also lead to young animal fatal myocarditis. Up to now, few researches foucus on FMDV myocarditis animal model and it's molecular mechanisms. In this study, the hearts of suckling mouse infected with FMDV were used as research objects. H.E staining of frozen sections and paraffin sections, transmission electron microscope technique, immunohistochemical analysis of FMDV and fibronectin (Fn), RT-PCR method for specific FMDV gene were used. Results indicated that suckling mice died 36h post FMDV infection, a lot of inflammatory cells infiltration in myocardium, mitochondria enlargement and myocardial cell lysis, sarcomeres disordered, FMDV replication occurred in heart and fibronectin(Fn) overexpressed in myocardial cell. Obvious pathological changes were not observed in the control group(PBS). Suckling mouse model for myocarditis after FMDV infection was established and it laid the foundation for research myocarditis molecular mechanism of young artiodactyl induced by FMDV.Gene expression of specific tissues in experimental condition can be detected fastly and comprehensively by high-throughput sequencing technologies digital gene expression profiling (DGE). At present, DGE was widely used in the field of drug research and development, medical research and basic science research etal. Researches of DGE in viral myocarditis was not found untill now. Our scientific questions are the following:which genes were differentially expressed in FMDV-infected heart and control group? What were the possible roles of these differentially expressed genes in FMDV-induced viral myocarditis? In order to answer these questions, DGE technology was used to analyze the differentially expressed genes between infected group and the control group suckling mice hearts based on above researches. Results showed that 3141 differentially expressed genes were detected including 2735 up-regulated expression genes,406 down-regulated expression genes. There were 47 differentially expressed genes were related to viral myocarditis pathway. Some of the 47 gene were validated by qRT-PCR and western-blot. Instruction and new ideas were provided by this study for futher research of FMDV-related myocarditis molecular mechanisms for suckling mice and other young cloven-hoofed animals.
