ObjectCD200is a glycoprotein widely expressed on lymphocytes, neurocytes as well as epithelial cells. Recent researches revealed that CD200-CD200R interaction plays an immunoregulatory role in inflammations. This study aims at the expression of CD200on apoptotic cells, especially the role CD200-CD200R plays in the cross-talking between the apoptotic cells and dendritic cells.MethodFirst the expression of CD200on both live cells and early apoptotic cells will be examined by flow cytometry, comparing SLE group with health control group. Then the peripheral monocyte derived dendritic cells and macrophages will be cultured in vitro, in order to count the contact ratio and engulfment ratio of different cell groups co-cultured with dendritic cells.Result1, In24h, the early apoptotic cells ratio is higher in SLE than in HC, while the patients with neuropsychiatric symptoms has distinctly lower ratio of early apoptotic cells.2, CD200expressed more on early apoptotic cells, comparing with live cells, and SLE patients has a more significant increase between the live and apoptotic cells.3, CD200rarely expressed on monocytes, while CD200R mainly expressed on CD14+cells, especially CD11c+cells.4, Early apoptotic cell has increased contact and inducing engulfment capability to DC, comparing with live cells.5, Live cells and early apoptotic cells expressing CD200have a tendency to keep DC away, while their counterparts not.ConclusionCD200express more on apoptotic cells than on live ones, but bearing CD200will gift the cells the capacity to keep the DC away. So CD200maybe play a role as " don’t eat me" signal. As long as PBMC from SLE patients express a more significant increase during its apoptotic stage, it may exaggerate the clearance deficiency of apoptotic materials, resulting in the overload of selfantigens in the circulation and causing the onset of SLE.
