BackgroundRNAlater is a non-toxic sample storage solution, which protects the sample RNA to ensure the downstream analysis. This study aims to investigate the role of RNAlater in long-term preservation of parathyroid tissue, and to provide a theoretical basis for the construction of tissue banks.Primary hyperparathyroidism, which is characterized by hypercalcemia, hypophosphatemia, bone lesions, and urinary tract stones is a heterogeneous desease in some cases and the diagnosis may be difficult using histological features itself. In2011, Giusti et al. found YWHAZ and VDAC1overexpressed in parathyroid adenoma than the normal glands. On the basis of this study, we aims to explore the expression of these genes in parathyroid adenomas in China, and to analyze the relationship between its expression and the clinical manifestations.Objective1. Role of RNAlater in protecting parathyroid adenomas samples is evaluated.2. Expression of YWHAZ and VDAC1in parathyroid adenomas and its relationship with the clinical manifestations are examined.Methods1.22parathyroid adenoma samples are collected,11are kept in RNAlater and the rest in frozen nitrogen. The expression of RNA between the2groups is compared.2.28parathyroid adenoma samples and clinical records are collected. Total RNA and protain are extracted. Realtime PCR and Weatern Blot are performed to evaluate the expressin level and its relationship with the clinical manifestations.Results1. RNAlater group has no advantage in the purity, concentration, and integrity of the extrated RNA, but has a higher expression level of the β-actin (P<0.01)2. The size of the adenoma is correlated to patients’total calcium, serum phosphate, ALP and PTH (P<0.05)3. The expression of YWHAZ is correlated to ionized calcium, total calcium, serum phosphate and degree of bone lesions (P<0.05)4. The expresse of VDAC1is low and shows no correlation to the clinical manifestations.Conclusions1. RNAlater has an advantage in protecting the RNA samples within2years 2. RNAlater shows a great advantage during the transportation of the samples in the domestic multicentre research3. Size of the adenoma has a positive correlation with the other clinical manifestations4. The expression of YWHAZ is correlated to the pateints’calcium and phosphate level, and is hopefully a new biomarker of parathyroid adenoma.5. The expression of VDAC1is low and shows no correlation to the clinical manifestations.
