Anti-effects On Acute Bacterial Peritonitis Of Total Glycosides From Paeonia Suffruticosa Andr. And Paeonia Lactiflora Pall

Objective:Extract total glycosides from Paeonia suffruticosa Andr.and Paeonia lactiflora Pall..Investigate the affects of them,which include the effect of antibacterial,promote blood circulation,protect small intestinal mucous membrane and decrease the leve of cytokine,in order to confirm drug effect substances and the anti-effects on acute bacterial peritonitis of them.Methods:Exp.1:Total glycosides were extract with 70%ethanol.Then use HPLC to quantitative the Paeoniflorin std.on the total glycosides.HPLC condition:separation column:Kromasil C₁₈(150 mm×4.6 mm,5μm),mobile phase:0.2%Phosphate buffer.MeOH(70:30),UV-VIS detector at 230 nm.Exp.2:Using E.coli,K.pneumonia,P.aemginosa and E,faecalis as tester strains to valuate the antibacterial effect of the total glycosides.The 2-fold agar dilution method were applied.Formulate 7 solutions whose concentration is half to the next one,then join into the culture medium,and bacterioagglutinin,35℃culture 18 hours.Exp.3.Animal model of acute bacterial peritonitis was established with cecal ligation plus puncture(CLP).The histomorphologic change of organs,the data of L/M, the level of cytokines were tested.Results:1.The content of total glycosides on Paeonia suffruticosa Andr.and Paeonia lactiflora Pall is 4%and 11%,the content of Paeoniflorin std.on them is 8.03mg/g and 23.5mg/g.2.The MIC of total glycosides to all bacterial species in our experients was been tested.The total glycosides from Paeonia suffruticosa Andr.have a a powerful effect on Pseudomonas Exotoxin,and the total glycosides from Paeonia Iactiflora Pall have a a powerful effect on E.coli.3.The total glycosides from Paeonia suffruticosa Andr.and from Paeonia lactiflora Pall both can promote blood circulation and decrease the leve of cytokine TNF-α;the total glycosides from Paeonia suffruticosa Andr.can protect small intestinal mucous membrane.Conclusion:1.The extract condition we selected is easy and convenient,and it make a high productivity.The analysis method we established is fit for testing the content of Paeoniflorin std.2.After testing the content of Paeoniflorin std,we confirm the 2 kinds of total glycosides have antibacterial activity to all bacterial species in our experiments.The total glycosides is one of active components.Compare with the studies we see before,our study data is accurate and actual,and provide a rational basis for clinical ues.3.After testing the antibacterial activity of the 2 kinds of total glycosides,we investigate if them have 3 anti-effects on acute bacterial peritonitis.And make sure them can promote blood circulation,protect small intestinal mucous membrane and decrease the leve of cytokine to provide a better rational basis for clinical ues.
