BackgroundLung cancer is one of the most common malignant carcinomas in China and even around the world. Because of its high degree of malignancy, it’s prone to proliferation, invasion and metastasis. While Cancer-derived exosomes play an important role in tumorigenesis, invasion and metastasis. Epidermal Growth Factor Receptor (EGFR) overexpression are involved in progression and metastasis of lung adenocarcinoma, but its relationship between cancer-derived exosomes and EGFR overexpression has not been studied too much.ObjectiveIn this study, we explored the effect of EGF on human lung adenocarcinoma cancer cell line and also examined whether EGFR associated proliferation promotion and metastasis promotion effects can be mediated by exosomes between cancer cells.MethodsUltracentrifugation-purified exosomes from the H322 cell line which were treated with EGF or not were studied by transmission electron microscopy(TEM), nanoparticle tracking analysis (NTA), Western blotting and measurement of exosomal protein concentration.The effects of EGF stimulated cancer cell-derived exosomes on the proliferation and migration of cancer cells has also been measured.ResultsUltracentrifugation-purified exosomes from the H322 cell line had a characteristic saucer-like shape and their diameter ranged from 40nm to 150nm. CD63 and TSG101 were abundant in exosomes. EGF had no effect on the morphology of exosomes, but stimulated their secretion.Lung adenocarcinoma cell-derived exosomes can significantly increase cancer cell proliferation, but there is no significant difference between EGF group and control group. While exosomes from EGF group can significantly increase cancer cell migration.ConclusionEGF has no effect on the morphology and particle diameter of lung adenocarcinoma-derived exosomes, but increase the secretion of exosomes.The EGFR associated migration promotion effect might be mediated by exosomes between cancer cells.
