Objective:Although SWNHs (single-wall carbon nanohorns) have been demonstrated to accumulate to cytotoxic levels within organs of various animal models and cell types, and it have been exploited for cancer therapies. The role of SWNHs in human glioma cell lines was unclear. To address this question, the research about direct role of SWNHs on the growth, proliferation and apoptosis of human glioma cell lines (U251) had been performed.Method:1. Different concentrate of SWNHs were used to culture human U251 cells. Microscope was employed to observe cell number,and cofocal microscope was facilitate to observe the morphology of cell and nucleus.2. The function of SWNHs on U251 cell proliferation and viability were detected by XTT assay and CCK-8 cell viability assay separately.3. Flow cytometry was used to observe the effects of SWNHs on apoptosis and cell cycle.Result:1. Our results indicate that particle size of SWNHs in water is between 342 to 712nm, the films of SEM show that SWNHs on PS surface are individual particles.2. SWNHs significantly delayed mitotic entry of human glioma cell lines cells, and inhibited its proliferation in a time and dose-dependent manner.3. SWNHs induced a significantly increase in G1 phase and inhibition of S phase followed the gradually increasing concentrations. SWNHs in human glioma cell lines cells induced significantly apoptosis followed bygradually increasing concentrations.4. The TEM images showed that individual spherical SWNHs particles smaller than 100nm in diameters were localized inside lysosomes of human glioma cell lines.Conclusion:SWNHs inhibited proliferation of human glioma cell lines, and promoted its apoptosis. Its specific mechanism may be through the Bcl-2 signal system, caused the U251 cells mitochondrial function obstacle. SWNHs may be as a novel opportunity or method for the research on treatment of human glioma.
