1. Research in tissue level and clinical analysis of B7-H3 and FOXP3 in primary human non-small cell lung cancerObjective:The study was designed to investigate the expression and relationship of B7-H3 and FOXP3 protein in primary human non-small cell lung cancer (NSCLC), and their correlation with clinicopathologic characteristic, for prove evidence for NSCLC of diagnosis and prognostic evaluation.Material and method:110 paraffin-embedded specimens were collected. The expression of B7-H3 and FOXP3 were measured by immunohistochemical (SP) methods. Analyze the relationship between the expression of B7-H3 and FOXP3 and the pathological characteristics, OS. The test and the Student two-tailed t test were performed to assess the relationship between expression of B7-H3, FOXP3+cells and clinicopathologic characteristic. Actuarial OS rates were calculated by the Kaplan-Meier method and analyzed using the log rank test. P values were considered to indicate a statistically significant difference at p<0.05. All analyses were performed using SPSS version 19.0 software (SPSS, Chicago,IL, USA).Results:1.There is no statistical significance difference between the positive rate of expression of B7-H3 and age, smoking index, primary tumor size, regional lymph node (P>0.05). The positive rate of expression of B7-H3 in male is higher than that is in female, the difference has statistical significance (P<0.05). The positive rate of expression of B7-H3 in Non-squamous NSCLC is lower than that is in female squamous NSCLC, the difference has statistical significance (P<0.01). The positive rate of expression of B7-H3 in TNM stage groupings I is lower than that in TNM stage groupings Ⅱ and III, the difference has statistical significance (P<0.05).2. There is no statistical significance between the positive rate of expression of FOXP3 and smoking index, primary tumor size, histology (P>0.05). The positive rate of expression of FOXP3 in≥60 year is higher than that is in<60 year, the difference has statistical significance (P<0.05). The positive rate of expression of FOXP3 in male is higher than that is in female, the difference has statistical significance (P<0.01).The positive rate of expression of FOXP3 in the lymph node metastasis group is higher than that is in the no lymph node metastasis group, the difference has statistical significance (P<0.05). The positive rate of expression of FOXP3 in TNM stage grouping I is lower than that in TNM stage groupings II and III, the difference has statistical significance (P<0.05).3. B7-H3 is positive correlation with FOXP3 in NSCLC (P<0.05).4.B7-H3 high expression is positive correlation with high percentage of FOXP3+Treg cells in NSCLC (P<0.05).5.Patients with B7-H3 high/FOXP3 high expression have shorter overall survival than patients with B7-H3 low/FOXP3 low expression (P<0.05)Conclusion:1.There were different expression of B7-H3 and FOXP3 in primary human non-small cell lung cancer (NSCLC)2. There is no statistical significance between the positive rate of the expression of B3-H7 and age, smoking index, primary tumor size, regional lymph node involvement in NSCLC, however there is statistical significance between the positive rate of the expression of B7-H3and gender, histology, neoplasm staging. There is no statistical significance between the positive rate of the expression of FOXP3 and smoking index, histology, primary tumor size in NSCLC,however there is statistical significance between the positive rate of the expression of FOXP3 and age, gender, regional lymph node involvement, neoplasm staging in NSCLC。3. B7-H3 is positive correlation with FOXP3 in NSCLC.4.B7-H3 high expression is positive correlation with high percentage of FOXP3+Treg cells in NSCLC (P<0.05).5. Patients with B7-H3 high/FOXP3 high expression have shorter overall survival than patients with B7-H3 low/FOXP3 low expression6.We first studied that the expression correlation of B7-H3 and FOXP3 and the significance with OS in NSCLC.2. The expression and clinical significance of immune related cytokines in peripheral blood of primary human non-small cell lung cancerObjective:According to the experimental results that B7-H3 is significant correlation with T cell, interleukin is detected to play an important role in the modulation of T cell,in order to study the expression of interleukin in peripheral blood of primary human non-small cell lung cancer, analyzing the relativity of NSCLC progression and prognostic evaluation.we choose IL-24, IL-36 as representor,using ELISA method to detect IL-24, IL-36 level of peripheral blood. The aim is to investigate the clinical significance of IL-24, IL-36, to prove evidence for NSCLC of prognosis.Method:We extracted serum samples from 10 healthy patients and 41 untreated patients with primary human non-small cell lung cancer. Changes of serum IL-24, IL-36 are detected using ELISA techniques, were compared with those in the gender, histology, the TNM stage.All analyses were performed using SPSS version 19.0 software (SPSS, Chicago, IL, USA).Results:1.The level of serum IL-24, IL-36 in non-small cell lung cancer patients was obviously higher than that in healthy subjects, and the difference between groups was significant (P<0.01)2.The level of IL-24 and IL-36 in peripheral serum of male patients was similar with that of female patients, and there was no difference between the two groups (P>0.05). The level of IL-24 and IL-36 in peripheral serum of male and female patients was higher with that in healthy subjects, respectively.4.The level of IL-24 in peripheral serum of NSCLC TNM stage I patients was higher than that of stage TNM stage Ⅱ-Ⅲ patients, and there was a difference between the two groups (P<0.05). The level of IL-24 in peripheral serum of NSCLC TNM stage I, Ⅱ-Ⅲ patients was higher than that in healthy subjects, and the difference between groups significant (P<0.01). The level of IL-36 in peripheral serum of NSCLC TNM stage I was lower than that of TNM stage Ⅱ-Ⅲ, and there was a difference between the two groups(P<0.05). The level of IL-36 in peripheral serum of TNM stage I patients was higher than that in healthy subjects, and the difference is between groups was not significant (P>0.01). The level of IL-36 in peripheral serum of TNM stage Ⅱ-Ⅲ patients was higher than that in healthy subjects, and the difference is between groups was not significant (P>0.01)Conclusion:1. The level of IL-24, IL-36 is different with the healthy patients in NSCLC patients.2. The level of IL-24 was gradually increased from in healthy patients, TNM stage Ⅱ-Ⅲ patients to in TNM stage Ⅰ patients, The level of IL-36 was gradually increased from in healthy patients, TNM stage Ⅰ patients to in TNM stage Ⅱ-Ⅲ patients.3. We first detected the significance of IL-36 in peripheral blood.4. We inferred there were positive correlation specifically between B7-H3 and IL-36 expression, negative correlation between B7-H3 and IL-24 expression.3. Verification of genes related to the non-small cell lung cancer metastasis. Objective:To study the effects of B7-H3 related to the local neoplasm metastasis and immune escape in the tumor, we studied multiple genes that are involved in the tumor progression in DNA transcription and regulation, in order to verify that the B7-H3 and FOXP3 in tumor induced process.Materials and methods:We extracted RNA samples from tumor tissue and paracancerous tissue of 10 subjects of untreated patients with primary human non-small cell lung cancer. We detected the expression of immune response effects transcription factor by real-time PCR, and analyze their correlation with B7-H3.All analyses were performed using SPSS version 19.0 software (SPSS, Chicago, IL, USA).Results:1. B7-H3 expression level in NSCLC tumor tissue of control group was higher than adjacent tissues, the expression level are different between the two groups (P<0.05), and TLR3 expression level in NSCLC tumor tissue are close to the control group. The expression of B7-H3 was no correlation with TLR3.2. TARK1 expression level in NSCLC tumor tissue was higher than adjacent tissues, the expression level are different between the two groups (P<0.05).The expression of B7-H3 were no correlation with TARK1 in adjacent tissues, but have possible correlation with TARKI in NSCLC tumor.3. EGFR expression level in NSCLC tumor tissue was higher than adjacent tissues, the expression level are different between the two groups (P<0.05).The expression of B7-H3 were no correlation with EGFR in adjacent tissues, but have possible correlation with EGFR in NSCLC tumor.4. TCF12 expression level in NSCLC tumor tissue are close to in adjacent tissues. The expression of B7-H3 was no correlation with TCF12.Conclusion:1 TRAK1 and EGFR expression in NSCLC was significantly higher than that in the control group (P<0.05), while TCF12 and TLR3 have closed expression levels with the control group.2 We first verified the correlation between B7-H3 with the relevant genes related with non-small cell lung cancer metastasis. As the experimental results, we inferred that B7-H3 expression was related to the expression of TRAK1 and EGFR in NSCLC lung cancer patients possible, and there were no significant correlation with TLR3 and TCF12.3. Above all, we predict that B7-H3 plays an important role in tumor progression with TRAK1, EGFR possibly.
