Objective:Liver cancer is a serious hazard to human health globally tumors. For Radix Bupleuri dispersing stagnated liver qi for relieving qi stagnation to medicine, and as a classic liver channel drug, is widely used in clinical treatment of liver cancer. This paper presents Radix Bupleuri has targeted to "modify" the anti-liver cancer drugs, which was delivered to the tumor site, in order to improve the therapeutic effect of this hypothesis. In this study, TG strains of flila-EGFP transgenic zebrafish as experimental object, with stereo fluorescence microscope as the main observation tool, objec-tive, systematic and direct evaluation of Radix Bupleuri collaborative Sorafenib efficacy of anti-HCC, and then verify that we propose the possibility hypothesis, provide objective experimental basis for treatment of Radix Bupleuri in clinical application in liver cancer.Methods:22hpf were observed in transgenic zebrafish embryo model vascular phenotype by a stereoscopic microscope (bright field) and stereo fluorescence microscope to evaluate the effect of Radix Bupleuri inhibiting angiogenesis. Record 2dpf wild type zebrafish embryos exposed to different concentrations Radix Bupleuri Water Extracts (RBWE) deaths, fitting LC50, LC10, eati-mate MNLC, and to evaluate the acute toxicity of RBWE. Con-struction of luciferase expression stably transfected HepG2 strains by their tumorigenicity in nude mice verification, by microinjection of CM-Dil staining reagent of stabletrans-fection strains injection into the 2dpf transgenic zebrafish embryo yolk sac, immediately assess human hepatoma transgenic zebrafish yolk sac tumor model is successfully established. On zebrafish embryos and larvae, the acute toxicity of Radix Bupleuri with light. RBWE induced wild-type zebrafish embryo mortality was a significant con-centration-dependent manner. RBWE LC50 of 72h of zebrafish embryo is 670 μg/ml, LC10 of 72h is 205 μg/ml,72h MNLC is 155 μg/ml. On zebrafish embryos and larvae, the acute toxi-city of Radix Bupleuri with light. The transgenic zebra yolk sac human hepatocellular carcinoma xenograft model was successfully established. Radix Bupleuri not have the effect of enhancing the inhibiting of sorafenib on transplanted tumor of human hepatocellular carcinoma growth in transgenic zebrafish.Conclusion:In the transgenic zebrafish model, Radix Bupleuri not have anti-angiogenesis effect.
