Investigation Of Pyoderma Cases Of Police Dog In Beijing Area And Etiological Analysis

The canine pyoderma is a commonly encountered disease which badly influences the working ability and efficiency of police dogs. Meanwhile, this zoonosis is also a risk for the personnel involved in training and management of police dogs. To understand the epidemiological status of the canine pyoderma in police dogs of Beijing area and the treating effect of lysin, the pyoderma cases occurred in one police dog base during October of 2013 to September of 2014 were analyzed, and 18 pyoderma samples and 150 healthy samples from different police dog bases located in Beijing were collected from October of 2014 to January of 2015 to detect the issue of Staphylococcus’drug resistance and bacteriostasis by phage lysin-SALy102.(1) The epidemiology research on the 47 pyoderma cases showed that, the pyoderma can occur in all seasons, with highest infectious rate in summer(40.12%). And the Springer Spaniel and German Shepard were two breads with highest mobility. The dogs at the age of 2 to 3 were more susceptible to the infection (over 50%). Meanwhile, the mobility of male dogs with pyoderma was a little higher than that of the female ones. To different body parts, infections in dorsal and inguinal areas were more often observed.(2) The pathogenic bacteria causing pyoderma was isolated from 18 infected dogs, which was identified by detecting the 16S rDNA, then the paper disk diffusion was used for testing its antibacterial sensitivity. The results showed that, staphylococcus pseudintermedius accounted 50% of the isolated bacterial, which were isolated from 94.4% samples collected from the ill dogs. In the antibiotics resistance test of staphylococcus isolated in these cases, the highest rate of drug resistant was to penicillin G at 50.56%, however methicillin-resistant Staphylococcus was not detected. And the multi-resistance rate reached 50%.(3) 150 samples from 50 healthy police dogs were collected on their body parts including periocular, dorsal and inguinal area.16S rDNA method and paper disk diffusion were both used for bacterial identification and antibacterial sensitive test for staphylococcus. The tests results showed that 15.33% samples contained staphylococcus. Staphylococcus pseudintermedius accounted for 26.1% in all over the staphylococcus. Considering the breeds, Springer Spaniel took the highest level of prevalence rate at 30.43% to staphylococcu. For the distribution at different body parts, the inguinal area involved at the highest rate of 26.09%. In terms of drug resistance test, only 26.09% of staphylococcus isolates were sensitive to drugs, which were even better than that of pyoderma samples.(4) Drug-resistant genetic screening was taken by PCR method as well.3 of 23 staphylococcus pseudintermedius samples were detected as negative in drug-resistant gene. In the drug-resistant genetic, linA/linA’ was with highest prevalence as 47.83%. The aacA-aphD and linA/linA’ took amount of 17.39% in all drug-resistant genetic typing. Combined with drug sensitivity test results, staphylococcus pseudintermedius in police dogs skin in Beijing area was with good drug susceptibility. The drug-resistant phenotype and drug-resistant genetic were basically consistent in these cases.(5) Lysin SALy102 inhibition and bacteria killing test were taken by MIC and OD variation. The MIC to staphylococcus aurous was 64-128μg/mL and that to staphylococcus pseudintermedius was 128-256 μg/mL. And the highest lysis efficacy was observed in 0-1 hours. Comparing with staphylococcus aureus, the staphylococcus pseudintermedius was more senetive to SALy102, but effective period was shorter.In conclusion, the study investigated the pyoderma cases of police dogs in Beijing and pathogenic analysis. It was preliminarily known of the regularity and incidence to pyoderma for police dogs in Beijing area, the antibacterial sensitivity pathogenic bacteria, as well as the lytic effect of lysin SALy102, which will provide theory basis for using the antimicrobials rationally and effectively and controlling the spread of drug-resistance pathogen.
