Objective: Study the relationship between ABCG2 and the arsenic-resistance of cell.Methods: (1) The expression of ABCG2 was detected in two kinds of induced arsenic-resisitant cells and bone marrow slices from 23 cases of acute leukemia patients. (2) The ABCG2 recombinants were transfected into 293T cell via lipofectamine 2000, after the expression vectors were transferred and identified by selection and sequencing, then the over-expression ABCG2 in 239T was detected with immuno-fluorescence stain and Western-blot. (3) The effect of over-expression ABCG2 to the proliferation of 293T cell was observed with Trypan blue stain and counting cells number. (4) The effect of over-expression ABCG2 to the arsenic-resistance of 293T cells was observed with MTT..Results: (1)The ABCG2 expression decreased in both two kinds of induced arsenic-resistant cells. (2) The expression level of ABCG2 protein in bone marrow of 23 cases of patients with acute leukemia were from low to high as follows:the normal control group, the first incidence of patients and the recurrence of the patient, The difference between the three groups was statistically significant (P <0.0001). (3) Over-expression of ABCG2 in 293T was comfirmed by immuno-fluorescence stain and Western-blot (4) Over-expression ABCG2 decreased the proliferative ability and arsenic–resistance of 293T cell.Conclusion: (1) Arsenic-resesistance of cells decreased the expression of ABCG2, and over-exoression of ABCG2 in cells would reduced arsenic-resesistance of cells. (2) The acute leukemia patients with high expression of ABCG2 would have a bad future; (3) ABCG2 gene over-expression cell model were successfully established.
