| Professor Guizhi Sun is a famous oncologist in the area of integrated Chinese and Western medicine. In her50years of clinical work, she has accumulated rich experience in treating various types of tumor patients from more than600,000person-times, and gotten outstanding curative effect, especially in the area of lung cancer treatment. For learning further academic thought of treating advanced lung cancer and further clinical experiences of preventing its recurrence and metastasis from the mentors, and exploring the material basis of its clinical curative effect, this article tried to summarize the theory, methods, prescriptions and medicines of the tutor about treating advanced lung cancer systematically under the careful guidance of the mentors, and tried to reveal the scientific connotation of Fuzhengpeiben rule of Chinese medical theory how to improve the quality of life and prolong the survival period of those patients with advanced lung cancer, and provide beneficial exploration contributed to clinical standardized treatment and further improve the curative effect of clinic practice.1Three Parts of the Thesis1.1Effective record of treating advanced lung cancer was analysized systematically by using the modern bio informatics.384prescriptions of31cases survived for more than three years in patients with advanced lung cancer have been analysized by the methods of constituent ratio analysis and scale-free network model resolution for a retrospective analysis to explore the characteristics of drug use and syndrome differentiation.1.2Academic thoughts of Professor Guizhi Sun in treating advanced lung cancer, and the treatment rules for preventing metastasis and recurrence of advanced lung cancer.Integrated ancient and modern literature, and combined with the principle and mechanism of those prescription and medicine, this article made a deep analysis about the etiology and pathogenesis of advanced lung cancer, and the therapeutic strategies, the characteristics of prescription and medicine, the rule of treatment for anti metastasis and recurrence, and the reason of significant effect.1.3Animal experiments have been completed to reveal the molecular and immunological mechanism of why Professor Guizhi Sun made remarkable clinical curative effect in treating advanced lung cancer.Su Qi extract is a representative prescription by which Professor Guizhi Sun has used in treating and preventing recurrence and metastasis of advanced lung cancer by the method of Yiqi Huoxue. Using the tumor bearing mice of Lewis lung cancer as model, the experiment would explore the molecular immunology mechanism of treatment done to advanced lung cancer and how the prescription effect on preventing the recurrence and metastasis, and interpret the scientific connotation of teacher Sun’s academic thoughts from a side.2main results2.1clinical studies2.1.1Study on the PrescriptionsQianjin Weijing decoction was used in a higher ratio in lung adenocarcinoma and small cell lung cancer than in lung squamous cell carcinoma (P<0.05); but there is no statistical difference between the pulmonary adenocarcinoma and small cell lung cancer (P>0.05).There is no statistical difference about Qingzao Jiufei decoction, Sanren decoction or Gualouxiebaibanxia decoction and combined Huangqi Jianzhong decoction and Baihe Gujin decoction amang those three kinds of lung cancer (P>0.05).Baihe Gujin decoction was used in a higher ratio in lung adenocarcinoma than in lung squamous cell carcinoma (P<0.05); but there is no statistical difference between the pulmonary adenocarcinoma and small cell lung cancer, the pulmonary adenocarcinoma and lung squamous cell carcinoma (P>0.05).Huangqi Jianzhong decoction was used in a higher ratio in lung squamous cell carcinoma than in lung adenocarcinoma (P<0.05); but there is no statistical difference between the pulmonary adenocarcinoma and small cell lung cancer, the lung squamous cell carcinoma and small cell lung cancer (P>0.05).2.1.2Study on the MedicinesAnalysized by scale-free network model, Professor Guizhi Sun is good at using small prescription such as Nidus Vespae and Pu Huang to detoxificate and anti cancer, Pangolin and turtle shell to drop off the node, Armadillidium and Bombyx Batryticatus and Jiuxiang pest to tongluo sanjie, and Diffusa and Paris polyphylla and Polygonum bistorta to clear heat and detoxication.The research also found that the combined Huangqi Jianzhong decoction and Baihe Gujin decoction was used the most widely amang those three kinds of lung cancer, and the Baihe Gujin decoction was the second one. On the other hand, the pathological change of excessive heat accumulation ocured in a higher frequence in the pulmonary adenocarcinoma and small cell lung cancer than in the lung squamous cell carcinoma. However, the pathological change of Yin deficiency and inner heat ocured in a higher frequence in the pulmonary adenocarcinoma than in lung squamous cell carcinoma, and the pathological change of Qi deficiency in lung ocured in a higher frequence in the lung squamous cell carcinoma than in pulmonary adenocarcinoma. This phenomenon may indicate that there were different pathological mechanisms in different kinds of lung cancer, and it need to be treat by different strategies.2.2Academic thoughts of treating advanced lung cancer and clinical experience of anti recurrence and metastasisProfessor Sun thought that there are several characteristics of advanced lung cancer for us to pay attention, such as the deficiency of righteousness Qi and excessive toxic heat, malignant sores and carbuncles, different pathological mechanism amang three different lung cancers, etc.The strategies of treating lung cancers should be considering those tumor as a sore without ulceration temporarily, and paying attention to replenish the Qi and blood in treating this kind of ulcer disease, combining the methods of disease differentiation and syndrome differentiation, treating them by integrated Chinese and Western medicine to achieve complementary advantages, etc. On the treatment methods, those lung cancer chould be mainly divided into six types. The heat toxin accumulation type chould be treated by Qianjin Weijing decoction. The heat injury body fluid type chould be treated by Qingzao Jiufei decoction. The heat injury Yin type chould be treated by Baihe Gujin decoction. The Qi deficiency type chould be treated by Huangqi Jianzhong decoction. The Qi deficiency with phlegm type chould be treated by Sanren decoction or Gualouxiebaibanxia decoction. And the Qi deficiency with Yin deficiency type chould be treated by combined Huangqi Jianzhong decoction and Baihe Gujin decoction.On the basis of prescriptions from disease differentiation and syndrome differentiation, some small prescriptions would be used to enhance the curative effect, such as pollen Typhae and nidus Vespae, Denon and silkworm, Armadillidium and Jiuxiang pest, inula flower and pumice, Platycodon grandiflorum and Flos farfarae, Pericarpium Trichosanthis and Allium Macrostemon and Pepper,The Preventive Treatment of Disease theory was used to guide the treatment of recurrence and metastasis of advanced lung cancer. At fact, the total pathogenesis of recurrence and metastasis of advanced lung cancer is still based on the deficiency of vital Qi and residual heat toxic. The prevention of recurrence and metastasis should pay attention to conditioning the body resistance function, coordination function of internal organs of the body to support healthy, enhance the body resistance, improve local tumor micro environment, inhibit cancer cell proliferation, etc.2.3Animal experimentThe experiment was aim to explore the mechanism of Su Qi extract effect on the Lewis lung cancer bearing mice from the regulation of Th17/Treg immune balance, and the influence of tumor inflammatory microenvironment and regulation of inflammatory factor activity and expression, and to discuss the molecular immunological mechanism of Su Qi extract against lung cancer recurrence and metastasis.The mouse tumor inhibition rate of cyclophosphamide group was higher than Su Qi extract group and Caesalpinia sappan group, and pulmonary metastasis inhibition rate of Su Qi extract group was higher than cyclophosphamide group and Caesalpinia sappan group, suggesting that Su Qi extract can inhibit the growth and metastasis of Lewis lung cancer, and combined with hematoxylin and Astragalus coulde enhance intervention effect higher than hematoxylin significantly (P<0.05).The splenic Th17and Treg cells of tumor bearing mice increased with time in tumor control group was significantly higher than those of the normal group (P<0.05), the treatment group decreased significantly than in tumor control group (P<0.05), and Su Qi extract group have significant difference than sappanwood group (P <0.05); reduced effect of Su Qi extract group on Treg cells is stronger than western medicine cyclophosphamide group (P<0.05), and the effects on Th17cells showed no significant difference with cyclophosphamide group (P>0.05); RORyt mRNA expression level and the level of Th17, Foxp3mRNA expression level and Treg level changed in the corresponding dynamic cell formation.In control group of tumor bearing mice, IL-17, IL-23, IFN-y, IL-4and IL-6were higher than that of the normal group, and showed different degree increasing, the medicine group after drug intervention and tumor control group had different degrees of decline; and in the tumor bearing mice control group, IL-2were lower than that of the normal group (P<0.05), after the intervention of the drug, the medicine group increased in different degrees than the tumor control group.According to the above results, it means that all drugs will inhibit the Lewis lung cancer metastasis, but the effect of Su Qi extract group was better than that of sappanwood group, and similar to cyclophosphamide group. Su Qi extract group was better than that of sappanwood group in regulating tumor inflammatory response, partial similarity to the Western medicine cyclophosphamide group, and part is slightly better than the western medicine cyclophosphamide group. The mechanism of Su Qi extract on Lewis lung carcinoma metastasis may be regulated the inflammatory microenvironment, inhibit local inflammatory response, enhance immune function. 3Main Innovation of this Research3.1Theory innovationAccording to the pathological features of tough, bad brittle and easily broken of the masses of advanced lung cancer, this article summarized the tutor’s academic thought in treating advanced lung cancer from sores theory and and her experience in anti recurrence and metastasis from four aspects, such as theory, method, prescription and medicine. It also revealed the etiology and pathogenesis, the strategy of treatment, the priciples of prescription to anti metastasis and recurrence. Research reveals the scientific connotation of the principle of Fuzhengpeiben in treating advanced lung cancer and anti metastasis, and provides a new idea for the deep study of academic thought.3.2Method innovationIn the process of excavating Professor Sun’s academic thought in treating advanced lung cancer, this study used modern biological information frontier technology, combined the analysis of the constituent ratio of dialectical prescription and scale-free network model organically, to mentor common dialectical prescription and drugs in the prescriptions, to reflect the experience of the famous TCM doctor exactly, and provide a methodology reference for the inheritance. There was no report of innovation relatively and it’s a method innovation.3.3Model innovation about inheriting the academic thought of famous TCM doctorIn the process of excavating Professor Sun’s academic thought in treating advanced lung cancer and preventing it from recurrence and metastasis, the outher edited and published a academic monographs, named Clinical Record of Sun Guizhi’s Experience of Treating Fifteen Kinds of Malignant Tumors.
