PurposeTo explore the association between the polymorphism of genes from renin-angiotensin-aldosterone systemand elite endurance athletes performance, and the mechanism of how the polymorphism works.MethodsAnalyze the polymorphism of22SNPs in12genes, such as AGT, REN, ACE, ACE2, AGTR1and so on, though MALDI-TOF, the differences of SNPs and haplotypes between atheletes and controls, and the assocations between polymorphisms/haplotypes and enduranceperformance. Build pcDNA3.1-AGTR1-T and pcDNA3.1-AGTR1-C plasmid, and transfect the plasmids into mammalian293T cells. Take pictures and detect mRNA levels after48hours.ResultsGenetype frequencis of AGTR1rs5182between athletes and controls, between top level athletes and controls, between5km runners and controls, and between10km runners and controls were significant differences. However,the differences of mRNA levels between genetype groups were not significant.Allele frequencis of ADD1rs4961and rs16843523and haplotypes frequencies of TT between10km runners and controls were significant difference. Haplotypes frequencies of CYP11B1and CYP11B2between top level athletes and controls, between marathon runners and controls and between female athletes and female controls were significant difference. But, further research is needed because the samples of10km runners,top level athletes,marathon runners and female athletes are small. The polymorphisms of AGT rs2148582, REN rs5707, REN rs11240688, ACE rs4344, ACE rs4343, AGTR1rs5182, AGTR2rs1403543are significant assocaiton with lung function. The polymorphisms ofAGTrs2148582, rs5051, RENrs11240688, ACErs4344, ACErs4363, ACE rs4343, NR3C2rs6810951and rs11737660, GNASrs7121are significant assocaiton with cardiovascular function. REN haplotypes, CYP11B1and CYP11B21-3haplotype, ADD1haplotypeare significant assocaiton with lung function. REN haplotype, ACEhaplotype, CYP11B1-CYP11B2(1-2and1-3) haplotypes are significant assocaiton with cardiovascular function.ConclusionAGTR1gene rs5182could be a candidategenetic mark of selectione lite endurance athletes in Han Population from Northern China, but this polymorphism does not affect aerobic capacity through changing AT1R mRNA levels. rs2148582、rs5707、rs11240688、rs4344、rs4343、rs5182、rs4363. rs6810951、rs11737660、rs7121、rs1403543, and REN、ACE、CYP11B1-CYP11B2、 ADD1haplotypes could be candidate molecular markers to predict lung and cardiovascular function of endurance athletes in Han Population from northern China.
