Objective:To investigate the expression of Nemo like kinase in colorectal cancer diagnosis andclinical application value.Materials and methods:In order to study the effect of NLK in rectal cancer development role, we collected406cases of colorectal cancer specimens, the expression methodin50cases of fresh frozen specimens using reverse transcription PCR,real-time fluorescence quantitative PCR and WestornBlot detection of NLKgene and protein in cancer tissue and normal tissue, and levels of expression by immune group detection of all NLK in the samples of this study, with the colorectal carcinoma and the correlation between survival rate.Results:(一) NLK in normal mucosa group, the expression of cancer tissues and lymph node metastasis group.Normal mucosa, colorectal cancer patients with lymph node metastasis group the positive expression rates of NLK were9.9%,57.6%,90.9%. The mean value of each group of data were compared, the overall level of NLKexpression was gradually rising trend, the difference was statistically significant between the3groups (P<0.001)(二) RT-PCR Westernblot NLK was detected in normal mucosa adjacent tothe carcinoma, expression(1) RT-PCRExpression of NLK mRNA in the adjacent normal tissue level (0.000094+.000082)×106copies/LDNA.The average expression of NLK mRNA in cancer tissues The level of (2.026667+0.624895)×106copies/L DNA,Significantly higher than that in the control group had significant difference(P<0.001)(2)The results of WesternBlotTo select five groups as shown in the Western blot results of50groups,can clearly see the tumor group protein bands are clearly visible, and the normal group bands dim or even no band, this shows that NLK has obviousexpression in colorectal cancer, and almost no expression in normal tissue.Comprehensive analysis of50groups of results showed that:the positive expression rate of NLK was66% in tumor adjacent normal tissues, the expression rate is18%, the difference wa statistically significant (P<0.01)(三) The relationship between NLK expression and clinical pathological factors in colorectal cancer(1) the expression of NLK and lymph node metastasis (P=0.001), distant metastasis (P=0), TNM staging (P=0) was positively related to;(2) NLK in low differentiation in colorectal cancer of the lower (P=0.019);(3) the level of NLK expression in rectal cancer was significantly higher than that of colon cancer (P<0.01).(4) the expression of NLK and sex, age, depth of invasion and differentiation degree of cancer has no obvious correlation(四) The relationship between NLK expression and survival rate of patients with colorectal cancer(1) survival rate was negatively correlated with the expression level of NLKand colorectal cancer patients overall survival (P=0),(2) single factor analysis showed that, the overall survival rate and TNM stage, tumor location, the expression level of NLK related, tumor free survival rate and TNM stage, the expression level of NLK related (all P<0.01)(3) in multivariate analysis, high expression of NLK and mucinous adenocarcinoma (P=0.029), rectal cancer (P=0), and low differentiation(P=0) and TNM stage (P=0) with correlation(4) the high expression of NLK and TNM staging (and disease-free survivalin patients with low (P=0) with correlationConclusion:the expression level of NLK can be used as importantpredictors of progression and prognosis of colorectal cancer, it has certainresearch value and application prospect.
