Objective:To clarify the suspended genetic and environmental risk factors of thrombotic diseases in Chinese Han population of Hunan Province, thus can provide theoretical basis for the prevention and treatment of thrombotic diseases.Methods:Case control study was performed in this study. We collected101cerebral infraction patients as CI group,73acute coronary syndrome patients as ACS group,59deep vein thrombosis patients as DVT group. Then we chose healthy individuals as control group, whose ages and sexes were matched with three case groups. We collected each individual’s medical history and recorded related examination result. Blood DNA Kit was used to extract DNA from venous blood. Then we used fluorescent PCR and SnaPshot methods to detect the polymorphisms of MTHFR C677T (rsl801133)、MTHFR A1298C (rs1801131)、MTRAsp919Gly (rs1805087)、CBS844ins68、FGB G455A (rs1800790)、F Ⅶ Arg353Gln(rs6046)、F ⅦHVR4、FⅫC46T(rs1801020)、THBDC1418T(rs1042579)、THBD G-33A (rs13306848)、 PROC Arg189Trp (rs146922325)、PROC c.574576delAAG (rs199469469)、PROCR Ser219Gly (rs867186)、PROS1A2148G (rs6123)、PAI-14G/5G (rs1799889)、t-PA C7351T (rs2020918)、ITGA2C807T (rs1126643)、ACE I/D (rs4646994)、APOE E2/E4(rs7412、rs429358)、 APOC3T455C (rs2854116)、PON1Arg192Gln (rs662).At last, we combined the environmental and genetic risk factors to analyse the related risk factors of thrombotic diseases.Results:1. The history of diabetes or hypertension, SBP, DBP, FBS, TG, HDL had significant differences between the CI group and the control group(P<0.05); the genotypic and allelotypic distribution of THBD C1418T (rs1042579) and THBD G-33A (rs13306848) had significant differences between the CI group and the control group (P<0.05); the allelotypic distribution of PAI-14G/5G and ApoE ε2/ε3/ε4had significant differences between the CI group and the control group (P<0.05). Logistic regression analysis showed that THBD G-33A AG genotype (OR=30.931,95%CI3.718-18.847), AA genotype (OR=8.372,95%CI6.309-151.652), history of diabetes (OR=3.464,95%CI1.442-8.320), TG (OR=2.504,95%CI2.090-3.001) may be independent risk factors of CI in Chinese Han population of Hunan Province. THBD C1418T CT genotype (OR=0.109,95%CI0.043-0.274), TT genotype (OR=0.017,95%CI0.001-0.258) may be independent protective factors of CI in Chinese Han population of Hunan Province。2. The history of diabetes, hypertension, drinking or smoking, SBP, DBP, TG, TC, HDL had significant differences between the ACS group and the control group(P<0.05); the genotypic and allelotypic distribution of THBD G-33A (rs13306848) had significant differences between the ACS group and the control group (P<0.05); the allelotypic distribution of PAI-14G/5G, ACE I/D had significant differences between the ACS group and the control group (P<0.05). Logistic regression analysis showed that THBD G-33A AG genotype (OR=21.688,95%CI6.861-68.560), AA genotype (OR=3.609,95%CI2.023-6.439), history of smoking (OR=6.039,95%CI3.420-10.666) and hypertension (OR-2.540,95%CI1.421-4.541) may be independent risk factors of ACS in Chinese Han population of Hunan Province. HDL (OR=0.052,95%CI0.018-0.151) may be an independent protective factor of ACS in Chinese Han population of Hunan Province.3. The HDL had significant differences between the DVT group and the control group (P<0.05); the genotypic and allelotypic distribution of THBD C1418T (rs1042579), THBD G-33A (rs13306848), PROC Arg189Trp (rs146922325) and PON1Gln192Arg (rs662) had significant differences between the DVT group and the control group (P<0.05); the allelotypic distribution of PROS1A2148G had significant differences between the CI group and the control group (P<0.05). Logistic regression analysis showed that THBD G-33A AG genotype (OR=34.924,95%CI7.489-162.864), AA genotype (OR=5.578,95%CI2.727-11.412) may be independent risk factors of DVT in Chinese Han population of Hunan Province. THBD C1418T CT genotype (OR=0.018,95%CI0.032-0.206), TT genotype (OR=0.081,95%CI0.015-0.450) and HDL (OR=0.018,95%CI0.005-0.063) may be independent protective factors of DVT in Chinese Han population of Hunan Province.Conclusions:1. The history of diabetes or hypertension, SBP, DBP, FBS, TG, HDL, THBD C1418T (rs1042579), THBD G-33A (rs13306848) may be risk factors of CI in Chinese Han population of Hunan Province; THBD G-33A AG/AA genotype, history of diabetes, TG may be independent risk factors of CI in Chinese Han population of Hunan Province. THBD C1418T CT/TT genotype may be an independent protective factor of CI in Chinese Han population of Hunan Province.2. The history of smoking, drinking, diabetes or hypertension, SBP, DBP, HDL, TC, TG, THBD G-33A (rs13306848) may be risk factors of ACS in Chinese Han population of Hunan Province; THBD G-33A AG/AA genotype may be an independent risk factor of ACS in Chinese Han population of Hunan Province. HDL may be an independent protective factor of ACS in Chinese Han population of Hunan Province.3. The history of HDL, THBD G-33A (rs13306848) may be risk factors of DVT in Chinese Han population of Hunan Province; THBD G-33A AG/AA genotype may be an independent risk factor of DVT in Chinese Han population of Hunan Province. HDL and THBD C1418T CT/TT genotype may be independent protective factors of DVT in Chinese Han population of Hunan Province.
