Objective:Gastric cancer (GC) is one of the most common malignancy and primary cause of death in Chinese cancer patients. Recurrence is a major factor leading to treatment failure and low level of5-year survival rate in GC patients following surgical resection. A major challenge for improving clinical outcomes is to recognize robust signatures for the prognosis of GC. The purpose of the present study is to identify a plasma microRNA (miRNA) that can serve as a novel prognostic biomarker and to assess its clinical applications in monitoring disease progression. In this study, we detected the expression of miR-191in gastric caner plasma samples using quantity real-time PCR and also studied the correlation between expression of miR-191and clinicopathologieal characteristics of gastric cancer. We aim to identify the role of miR-191in predicting recurrence of gastric cancer.Methods:In this study, we detected the relative expression of miR-191in gastric plasma samples of26patients, consisting of stageⅡ patients with recurrence and stageⅢ patients without recurrence, using real-time RT-PCR. We also analyzed the association between miR-191expression and clinicopathological characteristics in gastric cancer. Receiver operation characteristic(ROC) curve analysis was performed to determine the specificity and sensitivity of miR-191as a prognostic biomarker in gastric cancers.Results:The experssion of miR-191in plasma samples has remarkable differences in gastric cancer recurrence group and non-reccurrence group. miR-191has comparatively high expression in the gastric cancer recurrence group. High expression of miR-191was also correlated with depths of the invasion(p=0.021) and lymphatic involvement(p=0.014), but no relation with age, sex, histologic grade, vascular invasion, nerve invasion and adjuvent chemotharepy. The area under the receiver operating characteristic (ROC) curve of miR-191in differentiate GC recurrence and non-recurrence was0.913.Conclusion:Our results suggest that the miR-191as a predictor of disease progression has the potential to predict the recurrence risk for GC patients.
