| ObjectiveTo investigate the effect and mechanism of hyperbaric oxygen on intracephalic angiogenesis in rats with intracerebral hemorrhage for exploring the mechanism of hyperbaric oxygen in the treatment of intracerebral hemorrhage.MethodsThe rats were randomly divided into sham operation group (group A) and operation group, operation group which then randomly divided into intracerebral hemorrhage group (group B), hyperbaric oxygen group (group C). By Longa score, balancing beam score, Berderson score, body symmetry test score, we observe rat behavior change; Observation angiogenesis of brain tissue around hematoma in rats of each group by HE staining under the microscope; Using immunohistochemistry method, we measured the PCNA, vWF, HIF1-α, VEGF protein content in the brain of rats in each group; Using fluorescence quantitative RT-PCR method, we determined the expression level of HIF1-α mRNA, VEGF mRNA in brain tissue of rats in each group.ResultsBehavior score of experimental rats, A, B, C three groups in4d,7d,14d,21d and28d of the differences were statistically significant (P<0.01), hyperbaric oxygen group behavior score than intracerebral hemorrhage group decreased faster, significant differences between the two groups in14d,21d and28d were statistically significant (P<0.01).The HE staining was observed, a large number of vascular structure and capillaries can been seen in brain tissue around hematoma in intracerebral hemorrhage group and the HBO group.Expression of HIF1-α protein of tissue around the hematoma in the HBO group were higher than that in the intracerebral hemorrhage group, there was significant difference in14d,21d,28d between the two groups (P<0.01); Expression of PCNA protein of brain tissue around the hematoma in two groups in the corresponding time respectively, in the hyperbaric oxygen group were significantly higher than those in intracerebral hemorrhage group, in7d,14d,21d,28d difference between the two groups was statistically significant (P<0.05); the expression of VEGF protein in corresponding4d,7d,14d,21d,28d hyperbaric oxygen group were higher than that of intracerebral hemorrhage group, in14d,21d,28d difference between the two groups were statistically significant (P<0.01); The two group at the corresponding time respectively, the expression of vWF protein in the hyperbaric oxygen group were significantly higher than those in intracerebral hemorrhage group, in7d,14d,21d,28d difference between the two groups were statistically significant (P<0.01).The three group (sham operation group, intracerebral hemorrhage group, HBO group), the HBO group HIF1-α mRNA, VEGF mRNA expression values were higher than that of sham operation group and intracerebral hemorrhage group. In the14d,21d,28d difference of HIF1-α mRNA in the three groups were statistically significant (P<0.01). In the21d,28d difference of VEGF mRNA in the three groups were statistically significant (P<0.01).Conclusion1. Hyperbaric oxygen can improve neural function of rat with cerebral hemorrhage, promoting their rehabilitation.2. Hyperbaric oxygen may promote ICH-related angiogenesis by up-regulating the expression of HIF1-a and VEGF, to repair the damaged tissue. |
