Atherosclerosis is defined as a progressive, oxidative stress, inflammatory diseases, characterized in lipoprotein metabolism in the body, large and medium-sized artery intima dysfunction/apoptosis of endothelial cells, the accumulation of white blood cells and macrophages, infiltration, marked by a buildup of plaque. Blood mononuclear cells in the lesions of good hair area and endothelial surface adhesion molecules, in the role of chemokines, into the intima, and then into the macrophages, via a specific receptor mediated cell swallowing function accumulation of lipid, into a foam cells, line into the As early lesions.The doctor of traditional Chinese medicine thinks, atherosclerosis is associated with phlegm and blood stasis, mainly to with phlegm. Phlegm method widely used in the treatment of atherosclerosis and curative effect. To reflect the mechanism of the common law,"phlegm" lesions can’t focus on single medicine prescriptions and single experiment research, so the results may simply reflect the drug or the special role of the Chinese herbal formula. At the same time this study selects three representative prescription treatment phlegm, respectively is:two Chen Shang, fructus scallions white pinellia decoction and huanglian wendan tang, the above three-prescriptions as phlegm prescription is widely used in the treatment of atherosclerosis related diseases. Through the three phlegm prescription classified analysis of experimental results, find out the common parts, to reflect the common mechanism of phlegm treatment, thus further reflect the phlegm treatment by the pathological changes mechanism of common mechanism "phlegm". According to the above three prescriptions, this research is conducted in vivo and in vitro experiments in vivo and in vitro experiments this two parts.Trial In vivo:Objective:to study the three kinds of phlegm prescription apolipoprotein E knockout (Apo E-/-) mice aortic atherosclerosis effects and mechanisms.Methods:selects the ApoE knockout mice, feeding high fat forage duplicate atherosclerosis model, through three different phlegm prescriptons, testing its serum inflammatory molecules IFN-g, IL-6, IL-10, MCP-1, RANTES, VEGF and TNF alpha, and observe the change trend of aortic plaques, statistical analysis.Results:for atherosclerotic plaque area:fructus scallions white pinellia decoction group and huanglian wendan tang group compared to model group there were significant differences (P<0.05); For atherosclerotic plaque area correction:two Chen Shang, huanglian wendan tang group compared to model group had significant difference (p<0.05). Three kinds of phlegm prescription have different regulation effect on serum levels of inflammatory factors.Conclusion:the above phlegm prescription may through regulating different inflammatory factor levels and influence the Apo E gene knockout mice plaque area of atherosclerotic plaque, inhibit atherosclerosis disease process.Trial in vitro:Objective:to observe three phlegm prescription medicated serum on the effect of foamy macrophages.Method: choose THP-1human leukemia mononuclear cells, induced by PMA as macrophages, in three phlegm prescription medicated serum after join fresh ox-LDL copy AS foam cell model, oil red staining statistics, its bright red fat drop ratio, flow cytometry instrument statistics three serum anti apoptosis, THP-1observe the effect of three phlegm prescription on foamy macrophages and apoptosis.Results:huanglian wendan tang medicated serum could significantly inhibit the foamy macrophages, decrease the rate of apoptosis.Conclusion:hints that huanglian wendan tang atherosclerosis effect may be related to inhibition of macrophage phagocytosis of lipid and the associated macrophage apoptosis.
