[Abstract]Objective:To definite the difference of HSP70, IL-6, IL-8expression in rabbit VX2tumor after Radiofrequency ablation, and discuss the relationship between tumor RFA and immune responses.Methods:Tumor model was established by injecting VX2carcinoma cell suspension into the rabbit legs.30rabbits were divided into two groups:20for RFA treatment group and10for control group. The control groups only inserted electrodes, with simulation treatment. Blood samples were collected at5different time points, which were before and1st,3th,7th,14th days after RFA. The serum concentrations of HSP70, IL-6and IL-8were examined by ELISA (enzyme-linked immunosorbent assay). At each time point,4rabbits were executed in the treatment group, while2in control group. HSP70of tumor tissues was examined by immunohistochemical examination. PCR and Western Blot were used to detect the expression of HSP70and IL-8.At the same time, the tumor size was observed.Results:(1) After RFA, the growth rate of tumor slowed down.(2) The expression of HSP70in the remaining tumor tissues and in normal tissue adjacent to tumors increased,up to maximum in7days. But14days began to decrease. There was significant differences between the two groups (P<0.05).(3) The expression of interleukin8in the remaining tumor tissues and in normal tissue adjacent to tumors decreased after RFA(4) The expression of HSP70in the serum increased,up to maximum in7days. But14days began to decrease. There was significant differences between the two groups (P<0.05).(5). Interleukin8in the remaining tumor tissues and in normal tissue adjacent to tumors decreased in all rabbits of the two groups after RFA than that before the treatment (P<0.05), but there was no difference between the two groups.(6) Interleukin6increased in rabbits of the two groups after treatment (P<0.05), but It was lower in the RFA group than that in the control group. Conclusions:After partial treatment of RFA, the expression of Hsp70in serum, the remaining tumor tissues and in normal tissue adjacent to tumors was stimulated more efficiency, with IL-6in serum increasing, IL-8in serum,the remaining tumor tissues and in normal tissue adjacent to tumors decreasing. RFA may destroy the tumor tissue, release cytokines and tumor specific antigen, stimulating the body’s anti-tumor immunopotency.
