Glaucoma, as an irreversible blindness disease, is very difficult to be radical cure. After glaucoma patients been treated with long-term glaucoma drug and/or surgery, it is very common for them to get dry-eye related symptom. It has been documented that oral supplementation with ω-3essential polyunsaturated fatty acids could be effective to treat dry eye and some other chronic disease. So far, it is not routine treatment for dry eye or dry eye treatment after glaucoma treatment.In this project, we estimate ω-3essential polyunsaturated fatty acids effect on dry eye patients that under glaucoma treatment. By studying the sign and symptom change of the patient and tears cytokines expression level change, to explore possible mechanism in ω-3essential polyunsaturated fatty acids dry eye treatment.PART ONE Estimate ω-3essential polyunsaturated fatty acids effect on sign and symptom change in dry eye patient under glaucoma treatment.OBJECTIVE:Study ω-3essential polyunsaturated fatty acids effect on sign and symptom change in dry eye patient under glaucoma treatment.METHOD:103testees in our study has been grouped into (1) control group of healthy participants (CGH),(2) glaucoma on medication (GOM),(3) glaucoma after surgery (GAS), and (4) dry eye disorder (DED). In all these group, testee has been randomly get ω-3essential polyunsaturated fatty acids treatment for3months bid (+S) or sham drug (NS). In all the testees, symptom score, Schirmer’s test scores, luorescein tear BUT have been checked in3months follow-up.RESULT:1) in GOM+S and GAS+S group, symptom score decreased from6.83±3.74and9.78±4.87to3.25±1.71and4.04±2.03(P<0.05), while GOM+NS and GAS+NS group has no significant change2) Schirmer’s test scores, luorescein tear BUT in GOM+S and GAS+S group7.35±1.90mm and8.01±2.48s, increase compared to6.00±0.85mm and5.17±1.47s in GOM+NS and GAS+NS group (P<0.05). CONCLUSION:Oral supplementation with ω-3essential polyunsaturated fatty acids present a safe therapeutic advantage in patients with glaucoma suffering from dry eye disorders in sign and symptom.PART TWO: ω-3essential polyunsaturated fatty acids effect on tear cytokines change in glaucoma treatment induced dry eye.OBJECTIVE:To check co-3essential polyunsaturated fatty acids effect on sign and symptom change in dry eye patient that under glaucoma treatment.METHOD:103testees in our study has been grouped into (1) control group of healthy participants (CGH),(2) glaucoma on medication (GOM),(3) glaucoma after surgery (GAS), and (4) dry eye disorder (DED). In all these group, testee has been randomly get ω-3essential polyunsaturated fatty acids treatment for3months bid (+S) or sham drug (NS). We collected tear in all the testees in3months follow-up. Concentrations of specific cytokines in tears were determined by multiplexed particle-based flow cytometry.RESULT:1) In GOM, GAS, DED group, IL-1p\IL-2、IL-4、IL-6and TNF-α are all significant higher compared with CGH.2) After3months’treatment with ω-3essential polyunsaturated fatty acids, GOM+S, GAS+S, DED+S significantly decreased compared to GOM+NS and GAS+NS DES+NS in tear IL-1β level (P<0.05). GOM+S, GAS+S IL-6and TNF-a level significant decreased compared to GOM+NS and GAS+NS (P<0.05).CONCLUSION: ω-3essential polyunsaturated fatty acids could inhibit inflammation cytokines expression in dry eye patients.
