| In order to find a safe and effective treatment for colorectal cancer andfibrosarcoma using natural medicine, based on early study from our group, thisresearch project was designed as three main fields such as study on chemicalconstituents of the marine brown algae (ecklonia kurome Okama) includingextraction, separation and purification; chemical structure of phlorotannin PFFE-Ausing NMR technique methods; inhibition activity of PFFE-A on MMPs-1,2,3,9,13.The experimental results showed that PFFE-A is natural produced belong tophenolic compounds (phlorotannins) identified as an known compound; consideringthe unstable chemical properties of PFFE-A, this investigation systematicallystudied on its purification technology with very high recover rate, and due to thephenol hydroxy groups in this compound at multi-location and more numbers,therefore, for protection the chemical stability of the compound for activity process,this study provides practically experimental technology; meanwhile, the literatureresearch displayed, the compound has not been seen for the related research inbiological activities, however, the similar aggregate report showed that phenoliccompounds have bioactivity of multi-targeting and accordingly, hence, we havedesigned the study on molecular biological mechanisms of the system evaluation ofthe compound for fibrosarcoma cell in matrix MMPs-2,9series systems. The resultssuggested that MMPs well inhibited the biological activity of Expression againstMMPs, especially the overexpression of MMPs-2,9.To sum up, this conclusion can be drawn, namely the natural compoundPFFE-A significantly suppresses excessive expression of matrix Metalloproteinases, and based on the the fact for the natural source of PFFE-A easily and high yield,therefore, the further in-depth study of PFFE-A will provid a great potential newdrug with safe and effective properties for treatment of colorectal cancer andfibrosarcoma. While for the development of new drugs by inhibiting MMPssignaling factor mode this study will provide a scientific experimental evidence. |
