Coronary artery disease(CAD) is a main public health issue around the world. The pathogenesis is thought to result from an interaction between genetic background and environmental factors, Recent studies show that CAD may be closely related Angiotensinogen (AGT) gene M235T and T174M polymorsm. However, the results have been inconsistent, and there are still no relevant studies in our country.Objective:To investigated whether or not M235T and T174M polymorsm is the seperate risk factors, and the interaction of them and contaditional CAD risk factors have the synergistic effect.Methods:Genotyping of M235T and T174M will be detected with PCR methods in the Han population of Jingzhou,Hubei province.All CAD patients will be diagnosed by coronary angiography. The healthy people used as control. AGT gene M235T and T174M polymorphism will be detected by MALDI-TOF-MS. SHEsis soft will be used in genotype frequency calculation and haplotype analysis. The interaction of CHD and gene polymorsm will be analyzed by Logistic regression analysisResults:For M235T, the main genotype of patients and controls were TT(69.1%vs.59.5%). the main allele of patients and controls were T(59.2%vs61.3%). For T174M, the main genotype of patients and controls were TT(75.4%vs76.7%),The main allele of patients and controls were T(75.4%vs76.7%). M235T and T174M polymorphisms were in linkage disequilibrium. Haplotype Hap3(235T and174T) of AGT gene account for the highest proportion. Logistic regression analysis showed that M235T gene TT and MT genotype, alleleT, T174M gene TT, all Haplotypes are the sparated risk factors, ang have the interaction of other CAD risk factorsConclusions:AGT gene polymorphisms of Chinese Han population of Jingzhou has unique characteristics of distribution. M235T and T174M polymorphisms are the sparated risk factors. There is a synergistic effect on other CAD risk factors.
