| Objective To study the correlation of plasma phospholid transfer protein (PLTP) activity, PLTP rs2294213polymorphism and plasma sphingomyelin levels with lipids metabolism and coronary heart disease(CHD).Methods732subjects were divided into CHD (n=489) and control (n=243) groups by coronary angiography. Age, gender, BMI, history of smoking, hypetension, diabetes, lipid profiles (plasma TC, TG, HDL-C, LDL-C, apoA-â… , apoB, apoE and Lp(a)) and LVEF characterized by echocardiogram were obtained by clinical history in all subjects. Plasma PLTP activity, PLTP polymorphism (rs2294213) and plasma SM levels were measured and evaluated correlatively with lipid profiles and CHD.Results Plasma PLTP activity was significantly higher in subjects with ’CC’ polymorphism than that in those with ’GG’ and ’GC’ polymorphisms (26.7±12.8pmol/μL/hr vs.23.7±14.7/24.7±14.0pmol/μL/hr; p=0.04).Testing with an LSD test, subjects with ’CC’ polymorphism had significantly higher PLTP activity as compared to those with ’GG’ one (P=0.02). The proportion of CHD was not significantly different between PLTP genotypes (P=0.33). Based on univariate analyses, plasma PLTP activity was significantly associated with the presence of low HDL and PLTP genotype. In multivariable-adjusted analysis, low HDL and PLTP genotype remained independently associated with PLTP activity. In univariate linear regression analysis, PLTP activity was positively associated with plasma TG, apolipoprotein (apo)B and apoE, while it was negatively associated with plasma HDL and apoA-â… . Median SM levels were higher among patients with CHD than contrls (47.2vs40.7mg/dl, p<0.001). On univariate analyses, SM levels were significantly correlated with TG (r=0.10, p=0.005), apoB (r=0.13, p=0.001), apoE (r=0.135, p<0.001) and fibrinogen (r=0.11, p=0.006) levels and LVEF (r=-0.201, p<0.001). Using multivariate logistic regression analysis, SM levels were independently associated with the presence of CHD (aOR1.056, p<0.001).Conclusion (1)PLTP rs2294213genotype was independently associated with PLTP activity.(2) Human plasma PLTP activity was inversely associated with HDL levels and positively related to plasma apoB levels in this Chinese population.(3) The proatherogenic property of plasma SM might be related to CHD and abnormal metabolism of apoB-containing or TG-rich lipoproteins. |
