Protective Effects And Its Mechanisms On Semi- Bionic Extraction Of Compound Turmeric In Acute And Chronic Enteritis Rats

Abstract/Summary:
Compound turmeric is a prescription of Traditional Chiense Veterinary Medicine and has long been used for the treatment of acute or chronic enteritis, diarrhea, infectious fever, pneumonia, hepatitis, and common inflammation. The clinical effect of compound turmeric such as anti-inflammatory, antibacterial,and antipyretic analgesic was remarkable. However, the mechanisms of its effective in thetreatment of enteritis remain to bedefined.In this research, semi-bionic extraction of compound turmeric(SET) was perpared by using uniform design optimization. Parallel controlled trials were designed for ethanol extraction(AE), water extraction(WE) and semi-bionic extraction(SBE) of compound turmeric to determined the best extraction method by chemical and pharmacodynamic indexs.The aim of this work is to evaluated the acute toxicit, antipyretic effect, anti inflammatory and analgesic effects of SET, and investigate the protective effects of SET on acute enteritis(AE) and chronic enteritis(AE) induced by dextran sulfate sodium(DSS) in rats, to provide the basis for the development and research of compound turmeric. The main results were as follows:The extraction process of SET were studyed by uniform designe method. Using the contents of baicalin, berberine, emodin, curcumin, peony, gardenia, and the total flavonoids, alkaloids, alkaloids of SET to evaluationed the process parameters comprehensively. Combing the reality, the process parameters are determined as the extraction procedure of the first was 10 times the amount of water, p H 2.0, and extraction times were 120 min. The second was 6 times the amount of water, p H 7.0, and extraction times were 90 min. The third was 6 times the amount of water, p H 9.0, and extraction times were 90 min. The controllability and stability of method was better.Parallel controlled experiment were designed for AE, WE and SBE of compound turmeric to determined the best extraction method by chemical indexs which included the yield of dry extract, total anthraquinones, flavonoids and alkaloids,and by pharmacodynamic indexs, which included anti inflammatory(ear swelling in mice) effect and pharmacodynamic index(antibacterial test of Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Candida albicans in vitro). The results indicated that the order of extracts were SET>AE>WE, Prompted the technology was feasibled, and the semi-bionic extraction was superior to the traditional method.The acute toxicity of SET was demonstrated in preliminary experiments when administered to mice orally: LD50 = 4.20g/kg and 95% confidence limit 3.63-4.56 g/kg. SET has a significant inhibitory effect on acute inflammation by mouse xylene-induced auricular swelling and angle fork- induced foot swelling experiment.Effect of SET on body temperature of fever rats induced by yeast and induced by 2, 4-Dinitrophenol all shows SET has some antipyretic action and with dose effect relationship. Effects of SET on pain induced by acetic acid and induced by thermal stimulation in mice showed the certain analgesic effect.SET protects against DSS-induced in AE rats, which was dynamic, comprehensive in the present study. First, we monitored digestive function and morphology to better reflect the changes of AE progression at 1, 3, and 6 d. The findings showed that pretreatment with SET increased either the body weight or food intake in 24 h, indicating that SET protected gastrointestinal function in AE. We found that SET decreased significantly DAI and the colon W/D ratio, which indicates that SET could inhibit the leakage of serous fluid into intestinal tissue and attenuate the development of intestinal edema. In addition, SET significantly attenuated the increased IL-6, IL-8, and IL-1β production and also regulated the secretion of TNF-α in serum of rats with AE. The results suggest that pro-inflammatory cytokine production in serum was consistent with the severity of AE. In addition, TNF-α signaling affects various cytokines that also play a role in AE. In the present study, we also report that DSS caused a significant increase in the nuclear translocation of NF-κB-p65 in the colonic tissue 7 d after DSS challenge, whereas SET treatment significantly reduced NF-κB and ICAM-1 activation. In addition, we also found that pretreatment with SET significantly elevated the DAO and MDA activity in the colon and attenuated the increased MPO and GSH-Px production. The results support that decreased MPO and GSH-Px activity is associated with favorable prognosis of acute tissue injury. The results indicate that the protective effects of SET on AE induced by DSS were based on the improvement of the pathological changes in colon tissue, inhibition of NF-κB activation, and decreasing inflammatory cell infiltration and pro-inflammatory cytokine release.SET protects against DSS-induced in CE rats. CE rats were stimulated with the dilation balloon, SET significantly decreased AWR ratio at 21 d, 31 d, 41 d, and 51 d in high dose group, and at 41 d, 51 d in low dose group. The results suggested that SET could relieve the visceral pain sensitivity of CE induced by DSS in rats. ELISA assay showed that the serum levels of TNF-α, IL-6, IL-1, IL-8 and SET were significantly decreased, and the content of IL-10 was significantly increased. It also showed that the contents of MDA and MPO in colonic tissue were significantly decreased, and the contents of GSH-Px and DAO were significantly increased. The facts show that SET can be used as an effective therapeutic substance in CE.
