Studies On The Nanoemusion Preparation With Vitamin A,Oregano Oil And Allicin

In view of the serious infection of bacterial in broiler breeding and the drawbacks of antibiotic abuse, Origanum Oil, allicin and vitamin A (VA) were mixed to make into nanoemulsion preparation by the proper proportion. Furthermore, the pharmacokinetics, safety and the clinical application were studied, the objective was to develop a new type of antibacterial preparation, which had no toxicity and no residue and were expected to replace of antibiotics in the growing period, which had great importance to establish a bacterial infection control measure and green health care program. The main contents described as follows:(1)Preparation of the nanoemulsion of VA enriched with plant essential oil:VA and oregano oil, allicin were mixed as oil phase, with polyoxyethylene castor oil (EL) as surfactant, glycerol and ethanol as co-surfactant in the proportion of1:1while distilled water was used as the aqueous phase. Then compounded nanoemulsion was formed by using the pseudo ternary phase diagrams to determine the proportion of above components, as a result, the compounded nanoemulsion is a transparent red-orange liquid, the optimum Km value is1:1. The emulsion particle size had reached nanometer level measured by the potential Zeta diagram. In the preliminary stability test, the compounded nanoemulsion is stable in a certain temperature, ionic strength range, which had no change either by centrifugation or kept at room temperature for15days. Of all the antioxidants, the optimized composition of BHA and BHT were showed to have the good antioxidant effects on VA, which maintains more than95%residual in10days.(2) Pharmacokinetic of the compounded nanoemulsion:the compounded nanoemulsion along with different forms of VA, oregano oil and allicin were administered orally to the Haisaikesi layers, then blood were taken from the jugular fistula intensively at a Specific time points, and blood plasma were centrifugated and detected by HPLC. By comparing the peak time, peak concentrations and the medical area under the curve, lipid form as a reference form, compounded nanoemulsion had better absorption in blood cycles and higher bioavailability in comparison with a single component other two forms compared with a single form component (VA, oregano oil and allicin).(3) Safety test of the compounded nanoemulsion:Acute toxicity was Evaluated in mice, then LD50was1944mg of body weight; both the recommended dosage and3times of the recommended dose of the compounded nanoemulsion showed no toxicity to chicken, However,no significant difference was found in terms of blood index and main organ index could not cause blood index changes and organ damages, On contrary,a significant improvement of body weight was found in chickends administered with nanoemulsion.feed intake was found to be increasing in the birds with5-times dosage of nanoemulsion,no clinical signs were found.(4) Clinical application effect of the compounded nanoemulsion:The compounded nanoemulsion were Applied in3raising system (floor raising system,cage system and net floor system), compared with the differences between the experimental group and the control group. No statiscal difference was found in terms of feed takes,feed converted ratio,final body weight and survival rate. However,significant European benefit index was found in the nanoemulsion preparation.In summary, combination of VA with oregano oil and allicin proved to be a stable nanoemulsion preparation.In comparison with single prepation, it deserves a high dose in blood cycle and a good bioactivity.Moreover, it is safe for chicken application.With respect to protection, it proves to be effective both in vitro and in vivo.Clinically,it is able to improve broiler growth post administration.
