| Licorice flavonoids suspension was prepared by Licorice flavonoids extracts and excipients,in which Licorice flavonoids and Licochalcone is the main effective components.Objective: The pharmaceutical and pharmacodynamics studies on Licorice flavonoidssuspension will provide theoretical basis for the development of pilot production and clinicalapplication.Method: Pharmaceutical studies include the technical study, the establishment of qualitystandards and the stability study.Technical study: The optimal type and dosage of the excipients were explored bysingle-factor and orthogonal experiments.The quality standards: HPLC and ultraviolet spectrophotometry were used to determine thecontent of Licorice flavonoids and Licochalcone, respectively, which was verified by the methodsof the linear relation, stability, precision, specificity and recovery.Stability study: Acceleration test, accelerated testing light and long-term test assays wereemployed to determine the preservation conditions of Licorice flavonoids suspension.Toxicological evaluation: To evaluate toxicity and irritating of licorice flavonoids and itsexcipients, we did acute toxicity test in mice, chronic toxicity test in rats, muscle stimulation testand eye irritation test in rabbit.Pharmacodynamics study: The MICs of Licorice flavonoids suspension againststreptococcus suis and its growth curve treated with this compound were determined to evaluatethe bacteriostatic action of Licorice flavonoids suspension. The animal models of streptococcussuis infection was established and the clinical symptoms and pathological changes were observedto determine the protective effect of Licorice flavonoids suspension.Results: The results as follows:Technical study: The preparation method of Licorice flavonoids suspension: Heat themixture of hydrogenated castor oil and soy oil with heating electric stove. After adding soybeanoil into the colloid mill, while mixing the mixture with the added compounds later including thedissolved hydrogenated castor oil, Vitamin E, flavonoids extracts and glycerin monostearate while heating to distribute the compounds uniformly in the solvent. Finally, the rest of soybean oil wasadded.The quality standards: The results showed that the established methods were accurate,reliable and strong specificity.Stability study: This preparation should be preserved in a cool place away from light.Toxicological evaluation: This preparation is low toxicity, without irritation.Pharmacodynamics study: The MICs of Licorice flavonoids suspension for streptococcussuis were16μg/mL, which indicated that Licorice flavonoids suspension can inhibit the growthbacteria. The results of in vivo experiments showed that Licorice flavonoids suspensionsignificantly protect mice from streptococcus suis infection. Additionally, the cure rate of Licoriceflavonoids suspension for artificial infection swine streptococcosis was more than80%. |
