Sex gonad play an important role in maintaining animal reproductive system, secondary sex character and normal growth process. Ovariectomy comes a alteration in the appearance and abdominal fat deposited of hens, the mechanism of these changes is still unknown. Therefore, RNA-Seq was conducted to analyze the effect of ovarietomy on abdominal fat deposition from perspective of mRNA.20couples of Single Comb White Leghorn full sibs were divided into two groups,10hens in group1were ovariectomized at the age of9weeks and their intact sisters were taken as control. Another10hens were ovariectomized at the age of17weeks in group2. The same arrangement was carried out on Shouguang in group3and group4. The results showed that:1. The long and slender feathers were covered on the neck, rib and tail of ovariectomized hens. Ovariectomized hens had spur length approximately equal with normal male chicken (P>0.05). The comb height and wattle length of ovariectomized hens were significantly higher than intact hens but still lower than normal male chicken (P<0.05). We also found a hen in group4could crow.2. Ovariectomized hens had lower estrogen and higher testosterone concentration than intact hens (P<0.05).3. Testicle-like tissues were found in76.47%ovariectomized hens, among them,17.65%were at two sides,5.88%only at left side and41.18%only at right side, ovariotestis were found in11.76%ovariectomized hens.4. There were more abdominal adipose deposits in ovariectomized hens than intact hens (P<0.05).Livers of Single Comb White Leghorn in group2were chosen for further study. RNA-Seq analysis showed that267differential expression genes were screened by conditions of padj<0.05from all expressed genes.218genes, down-regulated in ovariectomized hens, were mostly enriched in Arrhythmogenic right ventricular cardiomyopathy, MAPK signaling pathway, Phosphatidylinositol signaling system, Hypertrophic cardiomyopathy, Glycerolipid metabolism, Glycerophospholipid metabolism and p53signaling pathway.49genes, up-regulated in ovariectomized hens, were enriched in Toll-like receptor signaling pathway, Prion diseases, Cytosolic DNA-sensing pathway, NOD-like receptor signaling pathway, Adipocytokine signaling pathway, Epithelial cell signaling in Helicobacter pylori infection, B cell receptor signaling pathway, Apoptosis. The results showed that the differential expressed genes mainly functioned on inflammatory and lipid metabolism.Genes, such as AGPAT2, PLD, PLCXD3, DGKE, DGKQ, FABP3, APOB and LPPR5, worked on phosphatidic acid synthesis, fatty acid esterification, apolipoprotein and phosphatidic acid hydrolysis were detected down-regulated in ovariectomized hens. GK5, functioned in glycerin phosphorylation, were more than2-fold up regulated in ovariectomized hens. What’s more, there was a correlation between immune response and lipid metabolism.Abdominal fat deposition related genes, which could be regulated by ovary, were identitied in this study and the research was helpful to lay the foundation for studies on poultry fat metabolism.
