Since the ancient times,people were being eager to live forever.They were trying to search for an elixir which could prolong life.Facts have proven that immortality has always been an elusive thing. However, when people play some correct and appropriate measures,such as sports,health food,skin care products and a variety of anti-aging medicine,et al.,They could effectively slow down the aging, reduce the occurrence of diseases associated with aging, improve the quality of life. At present,the world has entered an aging society.More and more scholars went into the study of aging and anti-aging.They were trying to find some drugs or methods which could delay aging or decrease the occurrence of diseases about old age. Kinetin (Kn), a type of cytokinin, is adenine derivative that promotes cell division, differentiation and growth.It has been involved in anti-oxidative damage and the powerful anti-aging effects. Kn can improve the activity of antioxidant enzyme such as SOD,CAT,GSH-PX ea al. and can be used as a direct free radical antagonist.Studies have proved that kinetin have obvious anti-aging effect on brain, liver, breast, skin, testis, ovary, heart and fruit fly larvae and pupae.Those prompted we thaut a question that whether the kinetin have effects of anti-oxidative damage and anti-aging.In this paper,Based on the above problems,this paper was designed on the free radical theory,mmune senescence theory, apoptosis theory.Objective:The purpose is to study the effects of Kn on immune function from morphology,cell structure, subcellular structure and gene.Methods:SD rats as the research object, Divided into the control group, aging model group and kinetin treatment group.The control group was injected subcutaneously with normal saline continuously for45days. Rats in other groups (aging rats, low Kn group, mid Kn group, high Kn group) were injected subcutaneously with D-galactose (125mg/kg-BW) to make aging rat models continuously for45days. In the process of experiment11d, rats in control group and aging model group were given normal saline while rats in other groups were given different doses (5,10,20mg/kg-BW) of kinetin continuously to45days.Duration of test,watching and recording the the morphological changes,the learning and memory ability statistical analysis of the immu of rats. Statistic analysisne immune organ index, the changes of cellular and subcellular structure;The change of antioxidant enzymes of immune organ; The changes of Serum immune factors IL-2, IL-6, immunoglobulin and T cell subsets; The change of erythrocyte immune function;The changes of△Ψm,apoptosis and apoptosis related gene Bcl-2, Bax mRNA of Immune organs.Results:(1)Aging rats droopy, thick coat, yellow,unresponsive, decreased activity, loss of appetite.Latentency shorten,error number increase, an effective shock; the index of Thymus and spleen decreased. Antioxidant enzymes s activity decreased uch as SOD,MDA content increased,the results indicated the aging rats model were success.(2) Kn could effectively improve the spirit of the aging SD rats induced by D-gal such as good appetite, smooth coat,the strong body,like activities,etc.Kn could improve the ability of the learing and memory such as the latency time extended gradually,fewer mistakes,the electric time is shortened obviously.Could increase the agility of the body, Avoid shock damage effectively.Could protect the cellular and subcellular structure of immune organs.Kn had the positive effect of delaying aging atrophy of immune organs.(3) Kn could improve the activity of antioxidant enzymes of immune organs.Kn could inhibit the free radicals accumulation,increased the content of IL-2and immunoglobulin,decreased the content of IL-6. The results support further the effect of Kn on anti-oxidative and anti-aging.(4) Kn could improve the stimulation indices of T and B lymphocyte.Could protected mitochondria from damage, maintaining high△Ψm.Decreasing the resting cell and increasing PI index, indicating that Kn can promote cell division and smooth cells from G0G1phase into S phase, which suggested that Kn could reduce the proportion of the resting cells,maintained vitality and immune ability of the spleen cell so as to protect against apoptosis and aging.Then protected against apoptosis by regulating the expression of Bcl-2and Bax expression.Conclusion:Kn could enhance the antioxidant enzyme activity of immune organs of aging model rats induced by D-gal.Could protect the structure of cells and subcellular of immune organs. Kinetin could regulate the expression of Bcl-2and Bax and effectively inhibit the apoptosis of spleenic lymphocytes and promote lymphocytes proliferation index so as to improve the body’s immunity and anti-aging effect.Change the content of immune factors and immune cells called T cells subgroup classification, improve the body’s immunity, delaying immunosenescence...
