Objective:To compare the variety of afferent information in nucleus tractus solitarii(NTS) of medulla oblongata by way of stimulus on Sibai,Dicang and Neiting of Foot Yangming Meridian,Quanliao of Hand Taiyang and non-acupoint,as well as gastric distention(GD),and to explore the special correlation between Sibai and the stomach by way of afferent information convergence in medulla oblongata's NTS.Method:In first experiment,Sibai,Dicang,Neiting,Quanliao and non-acupoint respectively were selected for body surface points stimulation,for which were given acupuncture,and extracellular recording method was applied to look for the neurons spike of NTS which had reaction from the above points stimulus, then the occur percent of reaction neurons(OPRN) in NTS(containing excited neurons and inhibited neurons) were calculated.In second experiment,firstly, extracellular recording method was used in NTS to validate GD related neurons which had excited and inhibited reaction when GD stimulus given.Secondly,in GD related neurons,Sibai,Dicang,Neiting,Quanliao and non-acupoint were given acupuncture respectively to look for the reaction neuron from above points stimulus,then the OPRN to body stimulus of NTS GD related neurons were calculated.In third experiment,immunohistochemistry was applied to compare the protooncogene c-fos expression in NTS induced by electroacupuncture(EA) at Sibai,Dicang,Neiting,Quanliao and non-acupoint, as well as GD stimulus.Results:1,In 52 rats,104 neurons spikes were recorded and 86 neurons with whole data.As for acupuncture at Sibai,the OPRN in NTS was 65.12%,and acupuncture at Dicang,Neiting,Quanliao and non-acupoint the OPRN were 50%,46.51%,34.88%,31.40%respectively.The statistics showed that after acupuncture at Sibai,OPRN was higher than that of acupuncture at Dicang, Neiting,Quanliao and non-acupoint respectively(P<0.01).And after acupuncture at Dicang and Neiting,OPRN was also higher than that of acupuncture at Quanliao and non-acupoint respectively(P<0.05).As for body surface points stimulation,the excited neurons in NTS prevailed.The excited neurons were selected,and their spikes change before and after body surface points stimulation were compared.For acupuncture at Sibai,Dicang,Neiting, Quanliao and non-acupoint,the neurons spikes change were(35.08±4.80)%, (28.25±5.46)%,(27.57±4.87)%,(20.02±4.23)%and(18.55±2.49)% respectively.There was significant difference among acupuncture at Sibai and others points stimulation(P<0.05 or P<0.01),and there was also significant difference between acupuncture at Dicang and Quanliao(P<0.01),Dicang and non-acupoint(P<0.01),Neiting and Quanliao(P<0.05),as well as Neiting and non-acupoint(P<0.01) respectively.2,In 52 rats,104 neurons spikes were recorded and in which there were 47 GD related neurons.In 47 GD related neurons,for acupuncture at Sibai,the OPRN in NTS was 80.85%,and acupuncture at Dicang,Neiting,Quanliao and nonpoint,the OPRN were 61.70%,57.45%,38.30%,36.17%respectively.The statistics showed that in GD related neurons,after acupuncture at Sibai,OPRN was higher than that of acupuncture at Dicang,Neiting,Quanliao and nonpoint respectively(P<0.01).And after acupuncture at Dicang and Neiting,OPRN was also higher than that of acupuncture at Quanliao and nonpoint respectively (P<0.01).In GD excited neuron,the excited neuron to body surface points stimulus were selected,and their spikes change before and after body surface points stimulus were compared.For those GD excited neuron,acupuncture at Sibai,Dicang,Neiting,Quanliao and non-acupoint,the excited neurons spikes change were(32.91±3.48)%,(26.79±4.16)%,(26.15±3.67)%,(21.04±2.69) %,(19.65±4.34)%respectively.There was significant difference among acupuncture at Sibai and others points stimulation(P<0.05 or P<0.01),and there was also significant difference between acupuncture at Dicang and Quanliao (P<0.05),Dicang and non-acupoint(P<0.05),as well as Neiting and non-acupoint(P<0.05) respectively.3,For the rats which were disposed by EA at Sibai and GD stimulus,there were more Fos-like-immunoreactivity(FLI) positive cells present mainly in medial subnucleus of NTS(mNTS),middle and caudal medulla oblongata prevailed (31.01±3.49,30.61±3.35 respectively).As for the FLI positive cells expression,there were no significant difference between EA at Sibai and GD stimulus(P>0.05),and there were significant difference between them and others(P<0.01 or P<0.05).There were some FLI positive cells present when EA at Dicang and Neiting(28.61±3.75,28.36±4.16 respectively),and there were significant difference between them and EA at Quanliao,non-acupoint and blank contrast group(P<0.01 or P<0.05).There were a few FLI positive cells present when EA at Quanliao(26.18±3.18),it was higher than that of EA at non-acupoint(14.76±3.06) and blank contrast group(5.50±2.30)(P<0.01). While there were few FLI positive cells present when EA at non-acupoint, which was still higher than that of blank contrast group(P<0.01).As for the blank contrast group,there were low density expression,when compared with others,there were very significant difference(P<0.01).Conclusion:1,Afferent information from body points stimulus could be recorded in NTS.2,The afferent information from body points stimulus which were recorder in NTS ranking from bigger to smaller were Sibai,Dicang,Neiting,Quanliao and non-acupoint,which suggest Foot-Yangming Meridian is more close to the Stomach.The afferent information were vary in NTS from the near and fast points of Foot Yangming Meridian,which suggest the Surface and Root theory of acupuncture have some science connotation.3,In kinds of doctrines which are used to explain body and internal viscera connection,meridians(acupoints) speciality could be explained well by the theory of meridians(acupoints) and vicera correlation,while couldn't be explained satisfied by modern medical science,therefore it is still deserved more research. |
