ObjectiveTo observe the influence of epimedium extraction on bone integrinαvβ3 expression in ratsMethods88 SD rats(female,260-300g)were randomly divided into control groups(12), pseudo-operation groups(16),model groups(12),positive groups(12),low dose epimedium groups(12),middle dose epimedium groups(12)and high dose epimedium groups(12).ovariectomy was performed in model,positive and epimedium groups,and rats in pseudo-operation groups were only cut on back and their ovaries were not excised.The control groups were not received operation.The medicine were given in day 7 after operation up to 12 weeks at once a day and the heavy and appetite of rats were measured weekly.The blood of rats were draw and centrifugated,then freezed in -80℃.the concentration of Ca and P ion,BGP, ALP,estradiol,corticosteroid and pregnancy hormone in serum were measured.Boen density,bone morphometry,bone integrinαvβ3 expression in femur and spine which were freezed at -80℃were measured.Results:Ca ion in high dose epimedium groups,P ion in high and low dose epimedium groups were significantly increased when compared with model groups.ALP activity in positive and all dose epimedium groups showed no significantly difference when compared with model groups(P>0.05),however,higher than in control and model groups.BGP in low dose epimedium groups were increased significantly than in model groups(P<0.05).BGP in high and middle dose epimedium groups were higher than in model groups but no significantly(P>0.05).Percent of bone trabecula area,number and resolving powder of bone trabecula in all dose Epimedium total flavonoids groups showed significantly difference (P<0.05).resolving powder of bone trabecula decreased,percent of bone trabecula area and number of bone trabecula increased.No difference were found in all dose Epimedium total flavonoids groups,percent of bone trabecula area and number of bone trabecula showed increasing pattern and resolving powder of bone trabecula showed decreasing pattern.After treatment 3 months,bone density of total femur in all dose Epimedium total flavonoids groups and positive groups increased significantly(P<0.05).bone density of total femur in control and pseudo-operation groups increased significantly than model groups(P<0.01)and no difference were found in all dose Epimedium total flavonoids groups,positive groups and control groups.We can indicate that bone loss in ovariectomy rats is severe,and Epimedium total flavonoids could increase bone density of femur.Era and Erβgene expression in model groups were lower than low and middle dose Epimedium total flavonoids groups(P<0.05),high dose Epimedium total flavonoids groups(P<0.01).αvβ3 gene expression showed same pattern but higher than other groups.Erαand Erβ3 gene expression in model groups were significantly lower than control group(P<0.05),andαvβ3 gene expression were significantly higher than contro groups(P<0.05).No difference were found in control and pseudo-operation groups.Conclusions1,Epimedium total flavonoids could treat osteoporosis in ovariectomy rats.2,Its mechanism is possible relate to increasing Ca,P ion concentration and ALP activity and evaluating BGP and estrogen level,moreover,decreasing integrinαvβ3 expression and then inhibiting bone resorption of osteoclast.Evaluating estrogen level and relating gene Era and Erβexpression were the major influencing factors.
