Background and Purpose Epithelial ovarian cancer is the leading cause of cancer death among women genital organs.Due to the asymptomatic nature and poor survival characteristic of the disease,screening and identifying specific biomarkers for ovarian cancer is an essential task.Methods In this study,proteomic methods were used to compare 6 pairs of the tumor and normal ovarian tissue samples derived from patients with epithelial ovarian cancer. With a chemically defined serum-free medium,the primary organ cultures were established from the paired tissue samples first.The condition media(CM) were then collected from the tissue cultures.Total proteins in the CM samples were separated by 10%SDS-PAGE,and then mass spectrometry(MS) was used to identify the proteins. A protein database of the condition media for ovarian cancer and normal ovarian tissue was set up and analyzed by the internet tools of DAVID Bioinformatics Resources, (http://david.abcc.ncifcrf.gov/home.jsp).Results Totally,1,413 proteins were identified in the condition media.Among those, 358 proteins were only detected in the conditional media of normal ovarian tissue,555 proteins were only detected in the conditional media of ovarian cancer groups.By DAVID Bioinformatics Resources analysis,it showed that secreted proteins and/or extracellular matrix proteins were over 20%.Functional analysis showed that in serum-free condition media of normal ovarian tissue,most proteins were secreted proteins which may play a role in the immune reaction and serine proteinase inhibitor. Most proteins were intracellular and cytoskeleton proteins in serum-free condition media of ovarian cancer which may take part in the process of cell adhesion or tumor progression and molecular chaperon.Conclusions With this approach,we discovered various secretory proteins associated with ovarian cancer in the serum-flee condition media.Data presented here provided a new way to analyze tumor markers of ovarian cancer in the peripheral blood and other kind of body fluids.It may help us to set up ovarian cancer proteomics for early detection and therapy...
