ObjectiveTo explore the pathogenic mechanism and therapeutic rules on acute myocardial infarction(AMI)from "collaterals blockage due to Qi deficiency and blood stasis" in theory.It was studied whether reinforcing Qi,remove blood stasis and dredging collaterals prescription have the effects of therapeutic angiogenesis in myocardium ischemic of infracted rats.And we have observed the morphologic changes and the expression of CD34,VEGF and bFGF at protein and mRNA level in myocardium of infracted rats to study myocardial angiogenesis and its molecular mechanisms.MethodsThe root of left coronary artery was ligatured to establish heart failure after myocardial infarction in wistar rats,and then it were divided into 6 groups at random and treated 2 weeks with different therapy.There are reinforcing Qi,remove blood stasis and dredging collaterals prescription group;Reinforcing Qi and remove blood stasis prescription group;Reinforcing Qi and dredging collaterals prescription group; Remove blood stasis and dredging collaterals prescription group;the model group;the sham operation group.14 days later,we observed their general status.Then we studied the expression of CD34,VEGF and bFGF at protein and mRNA level in myocardium of infracted rats,and measured micro vascular count(MVC),micro vascular density(MVD)in the edge of infarct section to determine if reinforcing Qi, remove blood stasis and dredging collaterals prescription could induce myocardial angiogenesis and evaluate its molecular mechanisms.Results1 Comparing of micro vascular density(MVD).It is difference that the model group were compared with the sham operation group in MVD(P<0.01).It is significant difference that the model group were compared with any other groups in MVD(P<0.01),and reinforcing Qi,remove blood stasis and dredging collaterals prescription group is differential than the reinforcing Qi and remove blood stasis prescription group(P<0.05)and the reinforcing Qi and dredging collaterals prescription group in MVD(P<0.01).The reinforcing Qi,remove blood stasis and dredging collaterals prescription group had more differential in MVD than the remove blood stasis and dredging collaterals prescription group,but it is no significant differential in MVD than the remove blood stasis and dredging collaterals prescription group(P>0.05).2 Comparing of the expression of VEGF at blockage due to Qi deficiency and blood stasis is the basic pathogenic mechanism of acute myocardial infarction(AMI),and Reinforcing Qi,remove blood stasis and dredging collaterals is its important therapeutic rules.The reinforcing Qi,remove blood stasis and dredging collaterals prescription possesses protective effects on experimental myocardial injury of infracted rats.It was proved that the reinforcing Qi,remove blood stasis and dredging collaterals prescription could promote angiogenesis and regulate up the expression of VEGF and bFGF in ischemic myocardium of infracted rats.It has been make sure that regulating of VEGF and bFGF is one of mechanisms of Therapeutic Angiogenesis by reinforcing Qi,remove blood stasis and dredging collaterals prescription.
