Influenza is an acute viral contagious respiratory disease caused by influenza virus,with serious symptom,high incidence rates,wide and rapid spread trend,now threats human health all the same.At present it is lack of effective prophylactic means.Adamantane and ribavirin are now widely used for viral disease,but their side effects are apparently.Influenza falls into the category of exopathic disease in Traditional Chinese Medicine(TCM).The traditional chinese medicine and Compound chinese Medicine have a great superiority in preventing and treating influenza,especially in dealing with symtoms and improving prognosis,now which is believed to be an important research way for the prevention and treatment of influenza.However,according to the variations of the body constitution and series of the disease,how to improve the effectiveness become a problem while treating with Chinese herbals.This article,including theoretic and experimental research,overviewed the development of Traditional Chinese Medicine(TCM)about the curing basyon the differentiation of symptom syndrome,and how to cure influenza virus using chinese herbals or decoction.Then the paper studied the way of treating influenza in basis of unification of "Differentiation of Six Channels" and "Differentiation of Four Systems(wei,qi,ying,and xue)" and "Differentiation of Tri-energizer".Then in order to study the mechanism and pharmacology ofⅠandⅡdecoction treating influenza,some experiments are did.PartⅠThe theoretic researehIn summary of the related contents in the medical and clinical experience of the medical practitioners,this article discusses the academic origin and developing routes of the exogenous febrile diseases in the related times from two aspects of the theories and therapeutic methods.《Plain Questions》put forward the concept of "febrile disease" and laid a theoretical foundation;《Classic on Medical Problem》pointed out an academic viewpoint of"five factor of febrile disease";《Treatise on Febrile Disease》created a therapeutic system of exogenous febrile diseases upon pattern identification;based on that,Doctrine of Epidemic Febrile Disease developed the theory of exopathic disease.What Treatise on febrile Diseases and Doctrine of Epidemic Febrile Disease discuss are both belonged to the category of exopathic diseases in Traditional Chinese Medicine.Actually,concerning about the TOM theory,"Differentiation of Six Channels" in Treatise on Febrile Disease and "Differentiation of Four Systems(wei,qi,ying,and xue)" and "Differentiation of Tri-energizer" in Doctrine of Epidemic Febrile Disease have similarities.The only difference among them is the expression of concepts,categories and terms when talking about theoriesMoreover,this article retropectively has given us a glimpse into several studies which include the research status quo of the pathogeny and epidemiology of influenza,the pathogenesis reseach of influenza and experimental research and progress of Traditional Chinese Medicine(TCM)in preventing and treating influenza,considered it had a wide prospect to treat and prevent the one with Traditional Chinese Medicine.under such a premise,studying the:Unification of Treatise on Febrile Disease and epidemic Febrile Disease" will be the beginning of the development of exopathic diseases in TCM.Then,this thesis integrate the theory of"Differentiation of Six Channels","Differentiation of Four Systems"(wei,qi,ying,and xue)and"Differentiation of Tri-energizer"to differentiate different syndrome of influenza,put forward that we should treat influenza according to different stage,and use medicine of both cold and hot natures simultaneously.The cause and pathogenesis of exopathic febrile disesae was ananlyzed,it was believed that the seasonal and latent pathogen was closely related to the health-qi.By clinical experience,it was stresses that the exopathic febrile disease,earlier or later,was of exterior-cold pattern.For Exterior syndromes,theⅠdecoction expell the pathogenic factors using pungent and heat or cold nature herbs,accompanied with cold nature herbs to clear excessive heat;For Interior syndromes,theⅡdecoction penetrate the pathogenic factors using pungent and heat herbs,clear excessivee heat and detoxicate with cold herbs,accompanied with resolving phelgm and blood stasis.PartⅡThe experimental researchTo study the mechanism ofⅠandⅡdecoction to cure influenza,we did some experiments.This experiments takes mouse infected by influenza virus dropping into the nose as the model and usesⅠandⅡdecoction to carry out experiments.The NIH mice is randomly divided into nine groups:normal control group,model control group,Ribavirin group,the group on high dose ofⅠ,the group on medium dose ofⅠ,the group on low dose ofⅠ,the group on high dose ofⅡ,the group on medium dose ofⅡ,the group on low dose ofⅡ.There are 10 mice in each group.Under slight anesthesia caused by ether,the mice are infected by dropping influenza vires into the nose.before the day the vires attacks,medicine is given.Then,according to the objective of every expeiment,relevant indexes are measured.The experiments included:1 Acute toxicity ofⅠandⅡdecoction【Objective and Methods】Using the method of modification KouShi,this experiment studied the toxicity ofⅠandⅡdecoction of high/medium/low dose in mouse.【Results】There is no mice died.【Conclusion】ⅠandⅡdecoction high/medium/low dose have no toxicity.2 The pharmacodynamics study【Objective and Methods】The pharmacodynamics studies are to confirm the effectiveness ofⅠandⅡdecoction.we observed the effects ofⅠandⅡdecoction on the lung exponent and life protection.【Results】Compared with model group,the death rate,lung exponent of the FM1 infected mice inⅠandⅡdecoction of high/medium/low dose groups all reduced obviously,the average life time of the FM1 infected mice inⅠandⅡdecoction of high/medium/low dose groups all improved obviously.Besides,the correlation of effectiveness and the medicine dose are direct ratio.【Conclusion】ⅠandⅡdecoction can alleviate the pathological changes in the lung of FM1 infected mice and have a protective effect on FM1 infected mice.3 The immune mechanism study【Objective and Methods】we use the method of ELISA to measure the inflammation correlatd cytokine serum content of FM1 infected mice,include TNF-α,IL-6,IFN-γ,IL-2,to explore the immune inflammatory mechanism ofⅠandⅡdecoction on influenza viral FM1 infected mice.【Results】Compared with normal control group,serum TNF-α,IL-6 content improved,IFN-γ,IL-2 serum content improved.compared with model control group,serum TNF-α,IL-6 content reduced,IFN-γ,IL-2 serum content improved.【Conclusions】ⅠandⅡdecoction inprove the immunological function,and have immunological regulation effect. 4 The Iipid peroxidation injuries study【Objective and Methods】We take the method of TBA and the method of purine oxidation to observed the effects ofⅠandⅡdecoction on the MDA and SOD content in the lung of FM1 infected mice,including differentiation phases 1th,3th,5th,8th,day.【Results】Compared with Normal control group,Model control group lung MDA content improved obviously,and SOD content reduced obviously;Compared with model group,ⅠandⅡdecoction of high/medium/low dose groups lung MDA reduced,SOD content improved.【Conclusions】ⅠandⅡdecoction could reduce endogenous toxin and improve the power of resisting toxin induced injury through reducing inflammation induced lipid peroxide and improve the power of resisting oxygen-derived free radidicals induced injury.5 Detecting the multiplication of influenza virus in the lung of FM1 infected mice【Objective and Methods】To study the antivirus effect ofⅠandⅡdecoction,We apply fluorescentreal-time reverse transcriptase-polymerase reaction in detecting influenza viruses.【Results】There is no copy of influenza virus in normal control group.The copy of influenza virus in the lung of model control group mice improved.compared with normal control group,the copy of influenza virus inⅠandⅡdecoction group are reduced.Compared withⅠdecoction group,the copy of influenza virus ofⅡdecoction group reduced.【Conclusions】This indicate thatⅠandⅡdecoction play an important in antivirus,and the antivirus effectiveness ofⅠdecoction is better thanⅡdecoction.
