The theory of Traditional Chinese Medicine(TCM) Zangxiang is the core of TCM theory system."Correspondence between man and nature"theory is the importance character of TCM. The standpoint of TCM which formed with the idea of Correspondence between man and nature theory is defer from biomedical model that instructed by modern western medicine reductionism. It stands out holistic harmonious control of human and form the theory of Correspondence between entrails and seasonal mode. It is the main different between TCM and western medicine that TCM know about human body from temporal organization angle and western medicine know about human body from spatial structure. It is the nature character of basic theory conception of TCM, such as"zang-xiang"theory, meridian theory. Accordingly, The theory of Correspondence between entrails and seasonal is an important approach of researching essence of"zang-xiang"from the angle of Correspondence between man and nature theory.1. ObjectiveThe study tends to reveal the essence of kidney in TCM conception from the angle of Correspondence between man and nature theory, combining melatonin seasonal secrete internal regulatory mechanism, taking melatonin as cut in point. Simultaneously, we introduced four season simulation system and artificially simulated natural season change. It provided a new way to research Correspondence between man and nature theory and"zang-xiang"theory. Thus, it provides a new idea to contemporary research work of TCM.2. Methods2.1 Theoretical methodsLiterature survey, logic analysis and other methods were used to analyze the necessity of researching Correspondence between man and nature theory using four season simulation system. We analyzed the feasibility and creditability of the research thinking of TCM kidney'essence taking AANAT as cut-in point and combining advanced research outcome of modern medicine. We raised a new way that use melatonin seasonal secrete internal regulatory mechanism to research the essence of kidney's temporal organization.2.2 Experimental methods2.2.1 Collectivity designTaking melatonin as cut-in point, we observed seasonal changes of serum MT, activity of pineal gland AANAT, protein level and RNA expression of rat in physiological state and analyzed the associability. Otherwise, we observed the same target in reversing seasonal state which made by the four season simulation system and compared with the natural state.2.2.2 Experimental animalSprague-Dawley male rats, weighted 180-200 grams, were randomly divided into groups as nature group, Simulation group. nature group : rats were adopted in light adequacy, temperature controllable and Natural light condition(room temperature was 17±2 degree in winter, it was 25±2 degree in summer). Simulation group: adopted four season simulation system to reverse season. The main Simulation factors were illumination, temperature and humidity.Illumination was taken as chief variable of season change. Rats ate and drank freely.Ratsdecollated at 9pm on winter Solstice and the Summer Solstice.2.2.3 Test methodsI125 -labeled radio-immunity for serum MT, isotope-labeled radio-immunity for activity AANAT, western blot for AANAT protein level, Real-time PCR for AANAT mRNA expression.2.2.4 Statistical analyses:Hormone levels were represented by mean±standard error (SE). Significance (p value) wascalculated with Duncan's multiple range test or Student's t test. The difference was significant when p<0.05.We used SPSS13.0 statistical package to analyzed difference between difference groups and used linear correlation analysis to analyzed variable relationship.3 Results3.1 In natural condition, activity of pineal gland AANAT, protein level and RNA expression presented a disposition tendency that it was high in winter and low in summer. Difference was significance. (p<0.05)3.2 In four season simulation system condition, activity of pineal gland AANAT and MT content in winter Simulation group was higher than summer Simulation group and was consistent with natural group. Difference was significance. (p<0.05). The difference between summer Simulation group and winter nature group and between winter Simulation group and summer nature group was statistical significance. But the difference between summer Simulation group and nature group and between winter Simulation group and nature group was not significance. (p>0.05) The data showed that using four season simulation system to do research work could be feasibility.There existed direct correlation between serum MT and activity of pineal gland AANAT, protein level and RNA expression in normal rat. Coefficient correlation separately were 0.950,0.722 and 0.659(p<0.01).In four season simulation system condition, it also had direct correlation. Coefficient correlation separately was 0.785,0.713,0.740(p<0.01)4 Conclusions4.1 Kidney regulation mechanism and seasonal secrete of MT and AANAT had close relationship. The essence of kidney's temporal organization had molecular biology groundwork.Our data showed that the function of serum MT, activity of pineal gland AANAT, protein level and RNA expression enhanced in winter and were direct correlation. Combining"kidney responds to winter"theory, we suggest that"kidney corresponds to winter"regulation mechanism and seasonal secrete of MT and AANAT had close relationship.4.2 It was a good way that known about complicated unitary regulation of organism and the essence of"zang"from temporal organization angle and"Correspondence between man and nature"theory.The essence of kidney's temporal organization had molecular biology groundwork. The influence of season to human body was not single regulation but came down to various layer of information regulatory mechanism. To a certain extent, it was a good way that known about complicated unitary regulation of organism and the essence of"zang"from temporal organization angle and"Correspondence between man and nature"theory.4.3 Four season simulation system could successfully simulate natural climate change approximately. It had feasibility and creditability to investigate"Correspondence between man and nature"theory by using the four season simulation system.Our data proved that four season simulation system successfully simulated environmental factors which were consisted four seasonal features, such as wind, rain, light, humidity, temperature. Four season simulation system successfully artificially simulated four seasons. Using four season simulation system to investigate had great feasibility and creditability. Searches proved that our study was advanced in international field.
