ObjectiveUnder the guidance of the theory of "Interactions Between Heaven and Mankind" in traditional Chinese Medicine,by observing the levels of-1(interleukin-1,IL-1),IL-2,IL-6,CD4+,CD8 +,IgE,IgG and TNF-? in the golden hamster in the four seasons and compared with the normal golden hamster,discuss the seasonal variation of various indexes of the immune system of golden hamster,and the golden hamster immune system rule of seasonal change of the indicators,and explore the pineal gland high regulating function of the body's immune system and the function in a " Interactions Between Viscera and Reasons "role,and further enrich the content of the theory of the five zang organs.Methods1.experimental animals:experimental animal for two months to the health of light sensitive male golden shrews(also called Syria to mice and hamsters),will be the golden mouse into the vernal equinox,summer solstice,autumnal equinox,winter solstice,four seasons group,35 days before each solar term purchase of experimental animals,golden gophers according to random number table method divided into experimental group(line the pineal gland excision technique)and control group(no treatment),experimental group and control group each 8,adaptive feed 3 days,in the vernal equinox day,the summer solstice,the autumnal equinox,30 days before the winter solstice four time points to start building experiments on animals.After the molds were made,the golden ground mice were normally kept until the vernal equinox,summer solstice,autumn equinox and winter solstice,and the animal sampling experiments were carried out after 9:00pm on the evening of the equinoxes and solstices.After weighing the golden hamster,under the dark room 5W red light,the abdomen aorta was used for blood extraction and spleen extraction after anesthesia.Put blood let stand 20 minutes at 4? in the centrifuge separation serum(3000 r/min,20 min),and spleen in-80? refrigerator frozen for later use.2.Detection method:ELISA was used to determine the content of IL-1,IL-2,IL-6 and IgE and IgG in the blood and IL-1,IL-2,IL-6,IgE,IgG,CD4+,CD8+,TNF-a in the spleen.3.data statistics and analysis:the experimental data with SPSS20.0 statistical software for data statistics,all the data for the measurement data,the data using the mean ±standard deviation(x + s)said,retain two decimal places,the experimental group and the control group between the two groups of data to test whether accord with normal distribution,conform to the normal distribution of date using independent sample t-test to compare,do not conform to the normal distribution of data using nonparametric test(two independent samples);The vernal equinox,summer solstice,autumnal equinox,winter solstice the data conforms to normal distribution between the four groups of the selection of single factor analysis of variance(one-way ANOVA)method,between two groups to compare with LSD method and T test,not normal distribution data using nonparametric test(k independent samples).The difference was statistically significant with P<0.05.Results1.The contents of IL-2,IL-6,IgE and IgG in serum of golden hamster had obvious seasonal variation.The content of IL-1,iIL-2,IL-6 and IgG reached the lowest in winter,while that of IL-1 control group reached the highest in autumn.The content of IL-6 in spring was significantly higher than that of other three seasons,and the content of IL-2 was the highest in summer.The content of IgE in the control group decreased continuously in spring,summer and autumn,and increased in winter.The content of IgG increased continuously in spring,summer and autumn,while decreased in winter,and there was a complementary relationship between IgE and IgG.2.The contents of IL-1,IL-2,IL-6,IgE,IgG,CD4+,CD8+ and TNF-a in the spleen of golden gerbil have obvious seasonal changes,which are characterized by high autumn and low winter.3.Data comparison between the experimental group and the control group showed that the pineal gland was involved in regulating the immune system,and some indexes lost the regularity of seasonal changes in the control group after the pineal gland was removed.Conclusion1.The immune function of the body has obvious seasonal changes,which is in line with the law of " Correspondence between human being and nature ".This study selected the immune indexes in blood present the summer solstice group of high levels of the vernal equinox,autumnal equinox the trend of the low level of winter solstice group,spleen disease index of the whole present the trend that higher autumnal equinox,winter solstice is low,the body's immune function during the rhythmic,and slightly down in winter.This phenomenon of adaptation changes of viscera function with seasonal change is one of the connotations of the theory of " viscera responding to nature" and the basic expression of the theory of "heaven and man responding".The seasonal variation of body function is closely related to the regulation of pineal gland.The pineal gland as the body of a kind of high regulating organs,to a certain extent to golden mice immune system play a regulatory role,the natural environment can affect the body function by the pineal gland,and adaptive response,is"Correspondence between human being and nature " play an important regulatory role in the process of tissues and organs,this is the pineal gland is the intermediary material provided the basis of" Correspondence between human being and nature".
