| PART â… INHIBITORY EFFECTS OF ANTI-CD137L ANTIBODY ON MURINE SGVHD AND AGVHDObjectiveBone marrow transplantation(BMT) is a major method to treat leukemia, malignant tumors, aplasia anaemia and immunodeficient diseases. BMT can be divided into autologous or syngeneic bone marrow transplantation and allogeneic bone marrow transplantation according to bone marrow sources. Autologous BMT and syngeneic BMT have the advantage of no obvious signs of rejection reaction due to their identical genotypes, but they have the disadvantages of relapsed leukemia and malignant cancers . Although allogeneic bone marrow transplantation can easily induce severe graft versus host disease due to HLA antigens incompatibility between donor and recipient and make the transplantation fail, they have good curative effect and less relapse in case that BMT succeeds.The data from clinical statistics demonstrated that patients sufferred from GVHD following allogeneic BMT have lower relapsed rate than similar patients without GVHD. It was confirmed in animal experiment that GVHD has graft-versus-leukemia (GVL) effects and can decrease relapsed rate of leukemia. These results suggested that one reason of leukemia recrudescence following auto-BMT is that auto-BMT has no GVL effect. If a GVL effect can be induced by artificial methods in patients following auto-BMT, the recrudescence of leukemia will be prevented. Glazier and Bryson reported that a GVHD-like syndrome, syngeneic GVHD(SGVHD),was developed following lethal irradiation, reconstitution with syngeneic bone marrow, and treatment with a 21-day course of the immunosuppressive agent cyclosporin A (CsA) in rat or mice, respectively.
