| AIM: To setup a rat allogeneic transplantation model to study the influence of myeloablative and nonmyeloablative preparative regimen on the levels of several serum Th1/Th2 cytokines and severity of aGVHD.METHODs: Our experiment established a rat allogeneic transplanta- tion model,donors were Wistar rats,recipients were SD rats.Forty receptor SD rats were randomized to four groups of A,B,C and D. A and B nonmyeloablative groups received total body irradiation(TBI) 2.0Gy. C and D myeloablative groups received TBI 6.0Gy.Four hours after TBI, rats of group B and D were injected with donor bone marrow cells via tail vein as test groups. Rats of group A and C were injected with saline as control groups.Afterward we observed severity of aGVHD and serum concentrations of IFN-γ,IL-2,IL-4,IL-10 measured by ELISA in different phases.RESULTs: After transplantation, groups of A,B and D rats regained their hematopoietic recovery on day(13.00±0.82),(13.10±0.88)and(13.70±0.95),respectively(P<0.05).Group C rats did not regain their hematopoietic recovery until died. The statistical difference of the median clinic aGVHD scores between group B and D was significant(P<0.05). According to the pathological evidence,group B rats hadⅡ-ⅢaGVHD and group D rats hadⅣ-ⅤaGVHD. The statistical difference of liver and colon pathological scores between group B and D was significant ( P<0.05 ) . Serum concentrations of IFN-γ,IL-2,IL-4,IL-10 in each group increased at different degrees, and levels of IFN-γ,IL-2 were significantly higher than IL-4,IL-10.Among all groups,peaks of cytokines in group D were the highest,and decreased in group B,C and A... |
