This scientific treatise is divided into 3 sections:The first section elaborated the effect on protien production and constituent of Malassezia Furfur in different culture conditions. By means of L16 (4~5) orthogonal design the first time, the different culture conditions of Malassezia Furfur (Pityrosporum Ovale)were determined . Factors to be observed included incubation temperature, incubation duration, vegetable oils and Tween , and four levels setting up for each factor. The protein content in the extracellular extract, cell wall extract and cytoplasm extract of Malassezia Furfur, which were obtained from different culture conditions, were detected with ultraviolet spectrophotometry, while the protein constituent of those were studied with SDS—PAGE. The optimal culture conditions for getting content the highest quantity and the most abundant constituent of protein in three extracts were obtained firstly. Our results showed that how high content of protein produced and how many constituent of proteins consisted of in the three extracts of Malassezia Furfur were closely related with five factors and four levels in our design; that all optimal culture conditions we have gotten by means of screening study were different in general, the optimal condition of the highest protein production in the extracts was at 30℃, 3.0 % glycerol monooleate, 2~7 days of incubation and sesame oil or olive oil overlaid; that the optimal conditions of the most abundant constituent of proteins in three extracts were mainly presence of sesame oil, and at 32℃ or 30℃ incubation for 4~7days. The condition that 37℃ incubation and 2% olive oil was adopted previously was not optimal one. Our study suggested that the culturing condition choice may paly an very important role in immunity research associated with Malassezia Furfur to be able to replicate anywhere and for any researcher.The second part dilineated the study on humoral immunity related with Malassezia Furfur of MFASD. Using case-control design and four antigenic preparations which included whole Malassezia Furfur(WMF), extracellular extract(ECE), cell wall extract(CWE) and cytoplasm extract(CPE), It was determined that classes of IgG, IgM, IgA antibodies against Malassezia Furfur in sera from patients with tinea versicolor, Malassezia Furfur folliculitis, seborrheic dermatitis, atopic dermatitis and four types psoriasis of MFASD with enzyme linked...
