Macrophage colony-stimulating factor (M-CSF) is an important member in the cytokine-regulating network. Due to alternative splicing, M-CSF can exist in at least three forms: soluble (s-M-CSF), membrane-bound (m-M-CSF) and extracellular matrix or proteoglycan (PG-M-CSF). High serum s-M-CSF level was reported in atherosclerosis, renal failure as well as pre-leukemia, leukemia, lymphoma and some solid tumors. Our previous work have shown that m-M-CSF and M-CSF-R were highly expressed in human leukemic cell line J6-1 and the proliferation of J6-1 cells depended on the auto-juxtacrine mechanism mediated by these molecules. In addition, the high expression of m-M-CSF and its receptor was detected on some malignant cells. In order to study the role of m-M-CSF and M-CSF-R in lymphoma, the expression of m-M-CSF and its receptor were detected on frozen sections from 12 Hodgkin's disease (HD) and 17 non-Hodgkin's lymphoma (NHL) lymph node samples by means of ABC immunohistochemistry. The results showed that the expression of m-M-CSF, PG-M-CSF and M-CSF-R could be detected in HD and NHL sections. In HD cases, RS or RS variant cells were positive stained. The positivity (75%) and intensities of positive reactions in HD samples were significantly higher than that in NHL cases. The positive reactions of both cytokine and receptor were observed in cell clusters while positive samples and positive cell clusters of them were identical. These results suggested that the expression of m-M-CSF, PG-M-CSF and their receptor be related to HD, and the juxtacrine mechanism exist in some lymphoma tissues, especially HD cases.In this report, we isolated M-CSF-like membrane associated growth factor receptor (MAF-J6-1-R) from J6-1 cell membrane by repeated freeze-thawing of J6-1 cells followed by sequential Sephadex G-100 and FPLC Mono Q chromatography, as well as recombinant human M-CSF soluble receptor (rh-M-CSF-sR) from E. coli. by His-Bind resin column.
