Objective:Recent studies show that cytokines play an important role in the development of SLE.The objective of this study is to detect serum levels of GM-CSF and GM-CSF Ab in patients with SLE and normal controls,and to analyze the relationship between SLE disease activity,C3,C4,24-hour urine proteinuria and serum GM-CSF and GM-CSF Ab levels,and to explore the possible roles of GM-CSF and GM-CSF Ab in the pathogenesis of SLE,especially with renal and blood injury.Try to find a special and sensitive biologic factor to help the diagnosis and treatment of SLE and to predict the prognosis,to provide clues to reveal the pathogenesis of SLE.Methods:The serum levels of GM-CSF and GM-CSF Ab were assayed in 40 patients with SLE,11 patients with other CTD and 20 healthy controls by ELISA.The differences in various SEE groups and the correlation with C3,C4,ESR,CRP, 24-hour proteinuria and the SLE disease activity of serum GM-CSF and GM-CSF Ab levels were researched.Results:.1.The serum levels of GM-CSF and GM-CSF Ab were significantly increased in SLE patients depending on the disease activity.The serum level of GM-CSF and GM-CSF Ab were significantly higher in patients with active groups than inactive SLE groups and normal controls,.Moreover,the serum levels of GM-CSF and GM-CSF Ab have positive correlation with the levels of serum level of IG-A,IG-M and SLEDAI(P<0.05).The serum level of GM-CSF and GM-CSF Ab have negative correlation with the levels of white blood cell(P<0.05).2.The serum level of GM-CSF and GM-CSF Ab were significantly higher in patients with active groups than inactive SLE groups and normal controls,and there were significant difference between patients with LN and no-renal involvement SLE group(P<0.05).3.There was correlation between serum levels of GM-CSF and GM-CSF Ab.Conclusions:1.The serum levels of GM-CSF and GM-CSF Ab were significantly increased in SLE patients depending on the disease activity and correlated with the SLEDAI score.The measurement of serum GM-CSF and GM-CSF Ab may be two useful indicators in SLE disease activity.2.The serum levels of GM-CSF and GM-CSF Ab were significantly descended after SLE treatment.,so serum levels of GM-CSF and GM-CSF Ab may give some directions to SLE prognosis.3.These results suggest that serum levels of GM-CSF and GM-CSF Ab combination with the clinical symptoms and the indicators of laboratory examination in patients with SLE maybe the useful tool for monitoring the disease activity,GM-CSF and GM-CSF Ab may be the drug targets of biotherapy in patients with SLE.4.GM-CSF and GM-CSF Ab may play a crucial role in the progress and development of SLE by the interaction of cytokine network...
