AP, JP and RPP patients differ in age and clinical features of periodontaldestruction These phenomena exhibit that different types of periodontitis have different pathogenies This study detected IL8 and TNFα in GCF and gingival tissues, observed the chemotaxis of polymorphonuclears and mononuclear cells of JP, RPP and AP, and tested the effect of Aa, Bm and Sp challenge on IL8 and TNFα production by HGF and HPDL1 the determination of IL8 and TNFα in GCF and gingival tissueGCF of AP, JP and RPP were collected and IL8 and TNFα were determined with ELISA method, and IL8mRNA and TNFα mRNA expression was detected by in situ hybridization The results showed that the positive rate of IL8 was 67.85% in AP GCF, while 34.48% in JP GCF and 25% in RPP GCF, the difference was significant between AP and JP or RPP group. The levels of IL8 were no difference in three groups The positive rate of TNFα in GCF were very low and without significant differences in three groups. IL8 mRNA and TNFα mRNA could be detected in the sites of gingval tissues where GCF were positive of IL8 and TNFα, while some sites where IL8 and TNFα were negtive in GCF also showed IL8 mRNA and TNFα mRNA expressions. IL8 and TNFα expression was observed in 1 of 4 healthy gingival tissues. Although it could be seen in the inflammated fibroblast tissues, IL8 mRNA was maily in the epithelium adjacent to the infecting microorganisms. TNFα expression was spread in all gingval tissue.
