Objective: To investigate highly efficient expression of imitative HPV16 L1 gene in prokaryotic system; To primarily investigate the application of serum anti-Ll protein IgG antibody in the monitor of cervical intraepithelial neoplasia.Methods: Constructed a recombinant plasmid pET30a(+)-L1(m202) which contained the imitative HPV16 L1 gene by gene recombinant technology. After its expression in E.Coli, the L1(m202) protein was purified and renatured. Anti-Ll protein antibody in patients with cervical neoplasm was detected by ELISA using denatured and renatured L1(m202) protein, respectively.Results: The pET30a(+)-L1(m202) plasmid was constructed successfully, HPV16-Ll(m202) protein was high-efficiently expressed in E.Coli and identified by SDS-PAGE and western blot. When denatured L1(m202) protein was used as antigen, 16.0%, 10.5% and 9.1% of the serum from patients with CIN> cervical cancer and controls exhibited positive reaction, and the positivity of HPV-DNA (+) CIN patients was 12.5%, there was no significant difference between the positivity of these three research group. When renatured L1(m202) protein was used, the positivity of patients with cervical cancer, CIN and control were 16.0%,35.5% and 15.9%, respectively, and the positivity of HPV-DNA(+) CIN pateints was 32.5%. The differences between CIN and control, CIN and cervical cancer, HPV-DNA(+) CIN and control were all significant.Conclusions: Anti-denatured L1 protein antibody in serum doesn't relate to cervical neoplasm. The serological assay of anti-L1 protein antibody using prokaryotically expressed renatured L1(m202) protein as antigen can detect the antibody produced after naturally HPV infection,...
