Mass Spectrometry In Drug Research

Some topics about the apllication of mass spectrometry in pharmaceutical sciceace were explored by means of GC/MS combined with other method such as MS/MS, direct inlet MS, HPLC and chemical synthesis. The thesis is composed of:1. Doping Control. The systematic GC/MS method was set up for detection of banned diuretics in human urine, through suitable derivatization of these polar and thermal-laibie compounds including methyiation, trimethylsillylation and extractive methylation. The overall method was highly sensitive and seletive, and had proved very effective in passing the accreditation test of International Olympic Committee and in the doping control of eleventh Asian Games in Beijijg.On the other hand, positive urine samples from volunteers were analysed. The metabolism of ethacrynic acid and spironolactone in human body were found to be different from those in publications.2. Mass Spectrometry. The fragmentation rules of over twenty diuretics were outlined by interpretation of the mass spectra of both free form and derivatives, and it was observed that derivatization may exhibit influences on fragmentation in many ways. Secondly, some new aspects of fragmentation mechanism of (?)carboline and p-methylphenylsulfonamide were proposed based on MS/Ms analysis of the deuterium lab(?)ed compounds. Thirdly, demethylation reaction of deoxyschisandrin was studied by deuterium synthesis and GC/MS.3. Essential oil analysis. Samples of essential oil extracted from several kindS of Chinese herb medicine were analysed by GC/MS and auto-computer spectra searching system. Some problems such as source development. variety defferentiation. and prescription study were discussed.
